Objective:To determine four lignans［podophyllotoxin-4-O-β-D-glucopyranoside(compound Ⅰ), 4'-demethylpodophyllotoxin(compound Ⅱ),podophyllotoxin(compound Ⅲ) and deoxypodophyllotoxin(compound Ⅳ)］ in Sinopodophyllum emodi Wall..Methods:The separation was performed with RP-HPLC on ZORBAX SB C₁₈ coloum (250 mm×4.6 mm,5 μm), using a mixture of acetonitrile-water as the mobile phase with gradient elution (0-13 min，25∶75；13-20 min，30∶70；20-30 min，32∶68；30-32 min，50∶50);The flow rate was 1.2 mL·min^-1 at 25 ℃,the wavelength for measurement was 290 nm.Results:The linearity was in the range of 0.05-5.0 μg(n=7) for compounds Ⅰ-Ⅳ, and all the correlation coefficients were higher than 0.9999; The average recoveries(n=6) for compoundsⅠ-Ⅳ were:102.2%(RSD=0.56%),101.0%(RSD=1.9%)，99.8%(RSD=0.87%)，102.6%(RSD=1.9%) respectively.Conclusion:This method is accurate to determine four lignans in Sinopodophyllum emodi Wall..
