Objective： To establish a HPLC analytical method for the Determination of content of buprenorphine in buprenorphine patchesand its skin permeation behavior in vitro. Methods ：The Symmetry used was the column(5μm, 3.9mm×150mm) with methanol-0.02mol·L-1 CH3COONH4(adjusting pH to 4.50 with NH3; 50:50, υ/υ) as mobile phase at flow rate of 1.0mL·min-1 and the detection was at 286nm.. Results ：The line range was 2.0 -10.0µg·mL-1(r=0.9999) with LOD of 0.2µg·mL-1. The precision of interday and intraday were less than 0.38 % and 0.98%, the average recovery was 99.9%~100.3%. Conclusion ：This method is accurate, reliable and simple and it is suitable for the determination of buprenorphine in buprenorphine patches.
