Abstract Objective: To determine the n-octanol/water partition coefficient and the solubility in various media of Gentiopicroside by HPLC.Methods: An HPLC method was established to detect the concentration of Gentiopicroside. The YWG ODS was used for separation column(10 μm，4.6 mm×250 mm )with n- octanol/water as mobile phase (1.0 mL·min-1) and detection at 270 nm.The partition coefficients for the n- octanol/water solution systems of Gentiopicroside were determined by shaking flask method. Results: The solubility of Gentiopicroside in water was 7.65 g/100 g and in n- octanol was 0.0578 g/100 g at 23 ℃; lg P of Gentiopicroside was -1.21. Conclusion: The HPLC method is simple and rapid to determine the oil-water partition coefficient of Gentiopicroside.
