摘要目的:建立猪苓药材中麦角甾醇的RP-HPLC含量测定方法。方法:猪苓经甲醇超声提取后,用HPLC测定。采用Diamosil C18(250 mm×4.6 mm,5 μm)色谱柱,流动相为甲醇 流速为1.0 mL·min-1 检测波长为283 nm 柱温30 ℃。结果:麦角甾醇进样量在1.64~16.4 μg范围内与峰面积呈良好的线性关系(r=0.9997) 麦角甾醇的平均回收率(n=5)为98.9%,RSD为1.2%。结论:采用RP-HPLC测定猪苓药材中麦角甾醇的含量,该方法灵敏、快速、准确,可作为2010年版中国药典一部增订猪苓含量测定项。

Abstract Objective:To establish an HPLC method for the determination of ergosterol content in Polyporus umbellatus by RP-HPLC.Method:Polyporus umbellatus was extracted with methanol by ultrasound extraction and the chromatographic procedure was carried out with Diamosil C18 (250 mm×4.6 mm,5 μm) column,The mobile phase was 100% methanol at a flow of 1.0 mL·min-1 and the detection wavelength was 283 nm.The temperature of column was 30 ℃.Results:The linear range of ergosterol was 1.64-16.4 μg (r=0.9997).The average recovery(n=5) of ergosterol was 98.9% and RSD was 1.2%.Conclusion:The method is simple,accurate and reproducible can offer references for the quality control of Polyporus umbellatus.
