芫花药材中4个黄酮苷元的定性鉴别定量分析___

摘要 目的:建立芫花药材中木犀草素、芹菜素、羟基芫花素、芫花素4个黄酮苷元的定性鉴别与定量分析方法。方法:采用薄层色谱法鉴别芫花药材中木犀草素、芹菜素、羟基芫花素和芫花素 采用高效液相色谱法同时测定这4个黄酮苷元的含量并制订含量限度要求。薄层色谱条件:以甲苯-乙酸乙酯-甲酸(8∶4∶0.2)系统展开,置365 nm下检视荧光。高效液相色谱条件:Kromasil C18色谱柱(4.6 mm×250 mm,5 μm) 流动相为甲醇-水(60∶40∶0.5),流速0.8 mL·min-1,检测波长350 nm(木犀草素、羟基芫花素)、338 nm(芹菜素、芫花素),柱温40 ℃,记录时间40 min。结果:芫花药材中木犀草素、芹菜素、羟基芫花素、芫花素4个成分的薄层色谱鉴别特征明显、专属性强。采用高效液相色谱法测定含量,木犀草素、芹菜素、羟基芫花素、芫花素的线性范围依次为0.032~0.16 μg(r=0.9994),0.072~0.36 μg(r=0.9995),0.098~0.49 μg(r=0.9995),0.15~0.75 μg(r=0.9997) 平均回收率(n=5)依次为99.86%(RSD=1.8%),101.0%(RSD=2.6%),100.3%(RSD=1.8%),100.7%(RSD=2.6%) 芫花药材中4种成分的总含量应不低于0.6%,其中木犀草素的含量不低于0.01%,芹菜素不低于0.2%,羟基芫花素不低于0.05%,芫花素不低于0.2%。结论:本方法操作简便,结果准确,能够同时测定芫花药材中4个黄酮苷元的含量,提高了芫花药材的质量控制标准,为其临床用药的安全性和有效性提供质量保证。

Abstract Objective:To develop qualitative identification and quantitative analysis method on 4 flavonoid aglycones of Flos genkwa.Method:Luteolin,apigenin,3′-hydroxygenkwanin and genkwanin were identified by thin-layer chromatography in Flos Genkwa.Toluene-ethyl acetate-formic acid (8∶4∶0.2) was used as developing solvent and the fluorescent of the compounds were obsevered under 365 nm.Their contents were determined by HPLC.Kromasil C18(4.6 mm×250 mm,5 μm) column was used.The mobile phase was methanol-water-glacial acetic acid(60∶40∶0.5) at a flow rate of 0.8 mL·min-1.The column temperature was 40 ℃.The detection wavelength of luteolin and 3′-hydroxygenkwanin were set at 350 nm,as well as apigenin and genkwanin were set at 338 nm.The record time was 40 min.Results:The linear ranges of luteolin,apigenin,3′-hydroxygenkwanin,and genkwanin were 0.032-0.16 μg(r=0.9994),0.072-0.36 μg(r=0.9995),0.098-0.49 μg(r=0.9995),and 0.15-0.75 μg(r=0.9997),respectively The average recoveries(n=5) were 99.86% with RSD=1.8%,101.0% with RSD=2.6%,100.3% with RSD=1.8%,and 100.7% with RSD=2.6%,respectively.By this HPLC method,the total content of four compounds should be equal or over 0.6%,and the content of luteolin,apigenin,3′-hydroxygenkwanin,genkwanin should be equal or over 0.01%,0.2%,0.05%,0.2%,separately.Conclusion:These analytical methods are simple,accurate with good reproducibility.Satisfactory results are obtained for the simultaneous determination of the four main flavonoid aglycones.There are suitable for quality control of Flos Genkwa.
