摘要 目的:建立准确、灵敏的HPLC-MS/MS方法鉴别中药制剂及保健品中非法添加的10种减肥药物。方法:采用UtimatesXB-C18色谱柱,梯度洗脱:流动相A为含0.1%冰醋酸的20mmol.L-1醋酸铵溶液,流动相B为甲醇,流速为1ml.min-1。选择正负离子全扫描方式检测临床常用的10种减肥药,利用质谱解析软件研究了上述化合物的质谱裂解规律,通过比较样品峰与对照品峰的一级质谱、二级质谱质荷比,确定样品中是否掺杂了化学药物。结果:各色谱峰间分离度良好,质谱分辨率符合要求,最小检出量为2ng至100ng。结论:该方法简便、快捷,灵敏度、准确性均可满足定性检查的要求。

Abstract Objective:To develop a HPLC–electrospray ion trap mass spectrometry for the identification of ten antiobesities illegally adulterated in traditional Chinese medicine(TCM) and Health foods. Method:The separation was conducted on a UtimatesXB-C18 Column by gradient elution with methanol and 20mmol.L-1 ammonium acetate as the mobile phase, the flow rate is 1ml.min-1. The mass spectrometer was operated in positive ion or negative mode and in three scan modes including full scan MS, selected ion monitoring and full scan MS2.The fragmentation pattern of these drugs was summarized,the obtained pseudomolecular ions, fragment ions were used to identify whether these drugs were present in the medicines. Results:Resolution of these ten drugs were good. The limits of detection were 2-100ng. Conclusions:The method is sensitive,specific,accurate and very applicable to Chinese Medicine and Health Food.
