RP-HPLC法同时测定小儿柴桂退热口服液中7个成分的含量

目的:建立同时测定小儿柴桂退热口服液中没食子酸、葛根素、芍药苷、黄芩苷、柴胡皂苷a、柴胡皂苷d和桂皮醛的含量测定方法。方法:采用反相高效液相色谱法,色谱柱为Thermo ODS C18(250 mm×4.6 mm,5 μm),以乙腈-0.1%磷酸水溶液为流动相,梯度洗脱,流速1.0 mL·min-1,检测波长254 nm(没食子酸、葛根素、芍药苷)、278 nm(黄芩苷)、210 nm (柴胡皂苷a、柴胡皂苷d)、290 nm(桂皮醛),柱温25℃,进样量20 μL。结果:没食子酸、葛根素、芍药苷、黄芩苷、柴胡皂苷a、柴胡皂苷d和桂皮醛检测质量浓度的线性范围分别为0.007 8~0.093 3、0.154 4~1.853 2、0.031 3~0.375 6、0.070 4~0.845 0、0.043 4~0.521 2、0.023 2~0.278 9、0.013 1~0.157 7 mg·mL-1r为0.999 4~0.999 9);平均加样回收率分别为101.1%、100.3%、98.8%、101.2%、98.3%、99.9%、99.4%(RSD<2.0%,n=6)。5批小儿柴桂退热口服液样品中测得的上述7个成分平均含量分别为0.200~0.221、4.146~4.200、0.818~0.856、1.698~1.754、1.086~1.236、0.586~0.732、0.084~0.320 mg·mL-1结论:该方法操作简便,结果准确,重复性好,可用于小儿柴桂退热口服液的质量控制。

Objective: To develop a method for simultaneous determination of gallic acid,puerarin,paeoniflorin,baicali,saikosaponin a,saikosaponin d and cinnamaldehyde in Xiao' er Chaigui Tuire oral liquid by RP-HPLC. Methods: The analysis was carried out on a Thermo ODS C18(250 mm×4.6 mm,5 μm)column with the mobile phase consisting of acetonitrile and 0.1% phosphoric acid in a gradient mode at a flow rate of 1.0 mL·min-1. The detection wavelengths were set at 254 nm for gallic acid,puerarin and paeoniflorin,278 nm for baicali,210 nm for saikosaponin a,saikosaponin d and 290 nm for cinnamaldehyde. The column temperature was set at 25℃ and the injection volume was 20 μL. Results: The linear rangs were 0.007 8-0.093 3, 0.154 4-1.853 2,0.031 3-0.375 6,0.070 4-0.845 0,0.043 4-0.521 2,0.023 2-0.278 9,0.013 1-0.157 7 mg·mL-1 for gallic acid,puerarin,paeoniflorin,baicali,saikosaponin a,saikosaponin d and cinnamaldehyde,respectively(r=0.999 4-0.999 9). Average recoveries were 101.1%,100.3%,98.8%,101.2%,98.3%,99.9% and 99.4%,respectively(RSD<2.0%,n=6). The average contents of the above 7 components in 5 samples were 0.200-0.221,4.146-4.200,0.818-0.856,1.698-1.754,1.086-1.236,0.586-0.732 and 0.084 -0.320 mg·mL-1,respectively.Conclusions: The established method is simple,accurate and reproducible. It can be used for quality control of Xiao'er Chaigui Tuire oral liquid.
