基于gyrB基因的伯克霍尔德菌属核酸测序鉴定与系统进化分析

目的:建立伯克霍尔德菌属典型菌株DNA促旋酶亚基B(gyrB)基因序列鉴定方法,为该菌属污染物的鉴定和分类提供研究思路和方法依据。方法:选择伯克霍尔德菌属内的典型菌株,以gyrB基因为研究靶点,设计引物,特异性扩增gyrB基因片段,测定核酸序列,并进行聚类比对分析。结果:伯克霍尔德菌属内的典型菌株鉴定的gyrB基因特征序列可满足"种"水平的鉴定。结论:本文建立了基于gyrB基因的伯克霍尔德菌属核酸测序鉴定方法,菌株鉴定的gyrB基因特征序列可满足"种"水平的鉴定,为该菌属污染物的鉴定与风险排查提供方法依据。

Objective: To establish the nucleic acid sequencing identification method for the typical DNA gyrase subunit B (gyrB) gene of Burkholderia, and provide research ideas and method basis for the identification and classification of Burkholderia pollutants. Methods: The typical strain in the genus of Burkholderia was selected, and the gyrB gene was used as a research target. Primers were designed to amplify the gyrB gene fragment, detect the specific nucleic acid sequence, and cluster alignment analysis was performed. Results: The characteristic gyrB gene sequence by typical strains in the genus of Burkholderia could identify the strain at the "species" level. Conclusion: In our study, a method for nucleic acid sequencing of Burkholderia based on gyrB gene is established. The gyrB gene signature sequence identified by the strain can satisfy the identification of "species" level, which is a typical Burkholderia contamination in quality control.
