新疆一枝蒿不同极性部位体外抗炎作用的谱-效关系分析

目的:研究新疆一枝蒿不同极性部位指纹图谱与体外抗炎作用的相关性,为揭示其抗炎作用物质基础提供依据。方法:采用高效液相色谱法建立新疆一枝蒿不同极性提取物及洗脱物的指纹图谱;以巨噬细胞RAW264.7为研究对象,考察新疆一枝蒿不同极性提取物及洗脱物对细胞上清液中一氧化氮(NO)、肿瘤坏死因子(TNF-α)、白介素-6(IL-6)、白介素-10(IL-10)和转录因子(NF-κB)的抑制作用,比较其抗炎活性差异;采用灰色关联度分析法建立谱-效关系。结果:新疆一支蒿不同极性提取物及洗脱物的指纹图谱有明显差异性,从HPLC指纹图谱中确定18个共有峰。体外抗炎结果显示,不同极性提取物及洗脱部位的抗炎作用存在显著差异,其中50%乙醇提取物的30%洗脱部位对体外炎症因子的抑制作用最强。谱-效结果分析,对药效贡献较大的峰为5个共有峰,分别为1、2、5、9、11号。结论:建立了新疆一支蒿的HPLC指纹图谱及其体外抗炎活性的谱-效关系,并确定了抗炎活性的主要有效成分,为该药材的药效物质基础研究提供参考。

Objective:To study the correlation between fingerprints and the anti-inflammatory effect in vitro of different polar parts of Artemisia rupestris L.,so as to reveal the material base of efficacy. Methods:HPLC was constructed to establish fingerprints of different polar extracts and eluates from Artemisia rupestris L. Using macrophage RAW264.7 as object,the inhibitory effects against NO,tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),interleukin-10(IL-10)and transcription factors(NF-κB)were investigated and anti-inflammatory effects of different parts were compared. The grey correlation analysis method was used to establish the spectrum-activity relationship. Results:The fingerprint of different polar extracts and eluates from Artemisia rupestris L. showed significant difference. 18 common peaks were determined. In vitro anti-inflammatory results indicated that the anti-inflammatory effects of different polar extracts and eluates were significant different. Among them,30% eluate from 50% ethanol extract had the strongest effect. Analysis of spectrum-activity relationship showed that 5 components had greater contribution to the effect,the peaks ranked as No.1,2,5,9 and 11. Conclusion:The spectrum-activity relationship of fingerprint and anti-inflammatory effect in vitro are established and the main anti-inflammatory components are identified,which providing reference for its study on therapeutic material basis.
