附子色泽与化学成分含量的相关性研究

目的:测定附子的色泽,建立附子中5-羟甲基糠醛(5-HMF)的含量测定方法,研究附子色泽与单酯型生物碱(苯甲酰新乌头原碱、苯甲酰乌头原碱、苯甲酰次乌头原碱)和5-HMF含量的相关性。方法:采用色差仪的△L、△a、△b和△E值测定和描述色泽,采用高效液相色谱法(HPLC法)测定3个单酯型生物碱、5-HMF的含量,采用SPSS软件的数理统计方法对色泽与3个单酯型生物碱的含量及其总含量、5-HMF的含量进行回归分析,确定色差值与化学成分的相关程度。结果:附子的色差值△E介于20.13~37.05,平均值为26.80;3个单酯型生物碱的含量分别介于0.001%~0.079%、0.001%~0.011%、0.001%~0.017%,5-HMF的含量介于0~0.002%;△E与苯甲酰新乌头原碱、苯甲酰乌头原碱呈显著正相关(P<0.05),与苯甲酰次乌头原碱及3个单酯型生物碱总含量呈极显著正相关(P<0.01),与5-hmf呈负相关。结论:传统的色泽描述可以采用色差值进行客观的测定,且这种色泽测定结果可以间接地反映其中化学成分的含量,对全面评价附子的质量提供了科学依据。

Objective:To determine the color of Aconiti Lateralis Radix Praeparata(ALRP),to establish a high performance liquid chromatography(HPLC)method for the content determination of 5-hydroxymethylfurfural(5-HMF)in ALRP,and to study the correlation between color and the contents of 3 monoester alkaloids(benzoylmesaconine,benzoylaconitine and benzoylhypacoitine)and 5-HMF in ALRP. Methods:The color was measured by colorimeter and described as values of △L,△a,△b and △E. The contents of 3 monoester alkaloids and 5-HMF were determined by HPLC. Regression analysis of color values and the contents of 3 monoester alkaloids and 5-HMF were performed by SPSS software. Results:The △E values of ALRP ranged from 20.13 to 37.05 with an average value of 26.8. The contents of 3 monoester alkaloids ranged from 0.001% to 0.079%,0.001% to 0.011%,and 0.001% to 0.017%,respectively. The contents of 5-HMF rangeed from 0 to 0.002%(not detected in some samples). There was a significant positive correlation(P<0.05)between △E values and the contents of benzoylmesaconine and benzoylaconitine and a highly significant positive correlation(P<0.01)between △E values and the contents of benzoylhypacoitine and the total contents of 3 monoester alkaloids. There was a negative correlation between △E values and the contents of 5-HMF. Conclusion:The traditional color description can be objectively determined by color values. The color values can indirectly reflect the contents of chemical constituents,which provides scientific basis for the comprehensive quality evaluation of ALRP.
