HPLC同步测定咳喘平散中4个成分的含量

目的:建立HPLC法同时测定咳喘平散中芥子碱硫氰酸盐、细辛脂素、五味子甲素、五味子乙素含量。方法:采用C18(4.6 mm×250 mm,5 μm)色谱柱,流动相为乙腈-0.08 mol·L-1磷酸二氢钠水溶液,梯度洗脱,流速1.0 mL·min-1,柱温30℃,检测波长为286 nm(芥子碱硫氰酸盐、细辛脂素)、220 nm(五味子甲素、五味子乙素)。结果:芥子碱硫氰酸盐、细辛脂素、五味子甲素、五味子乙素线性范围为0.238~1.789、0.061~0.457、0.071~0.533和0.069~0.521 μg;平均回收率为98.7%~101.8%,RSD为1.6%~1.8%。3批样品中芥子碱硫氰酸盐、细辛脂素、五味子甲素和五味子乙素4个成分的含量分别为1.508~1.566、0.470~0.497、0.398~0.420、0.470~0.553 mg·g-1结论:本法可为咳喘平散及其类似组方制剂的质量评价提供研究基础。

Objective: To establish an HPLC method for determination of sinapine thiocyanate, asarinin, deoxyschizandrin and schizandrin B in Kechuanping powder.Methods: A ZORBAX SB-AQ C18 column (4.6 mm×250 mm,5 μm)was used,the mobile phase consisted of acetonitrile-0.08 mol·L-1 sodium dihydrogen phosphate aqueous solution with gradient elution at the flow rate of 1.0 mL·min-1.The column temperature was set at 30℃,and multiple detection wavelengths were set at 286 nm for sinapine thiocyanate and asarinin and 220 nm for deoxyschizandrin and schizandrin B.Results: The linear ranges of sinapine thiocyanate,asarinin, deoxyschizandrin and schizandrin B were 0.238-1.789 mg,0.061-0.457 mg,0.071-0.533 mg and 0.069-0.521 μg,respectively;the average recovery rates were 98.7%-101.8%,RSD were 1.6%-1.8%.The contents of above 4 components in 3 tested samples were in the ranges of 1.508-1.566 mg·g-1 for sinapine thiocyanate, 0.470-0.497 mg·g-1 for asarinin,0.398-0.420 mg·g-1 for deoxyschizandrin and 0.470-0.553 mg·g-1 for schisandrin B in Kechuanping powder.Conclusion: The study provides scientific basis for the quality evaluation of Kechuanping powder and preparations of the same kind.
