HPLC法同时测定漏芦花中6个成分的含量

目的:建立HPLC法测定蒙药漏芦花中槲皮素-3-O-α-L-鼠李糖苷、木犀草素、芹菜素、5,7,4'-三羟基-3'-甲氧基黄酮、蜕皮甾酮和泥胡木脂素B葡萄糖苷的含量。方法:采用YMC-Pack C18色谱柱(4.6 mm×250 mm,5.0 μm),以乙腈(A)-0.2%磷酸水溶液(B)为流动相,梯度洗脱,流速0.8 mL·min-1,柱温35℃,检测波长254 nm。结果:槲皮素-3-O-α-L-鼠李糖苷、木犀草素、芹菜素、5,7,4'-三羟基-3'-甲氧基黄酮、蜕皮甾酮和泥胡木脂素B葡萄糖苷6个成分的进样量分别在0.090~2.24 μg(r=1.000 0)、0.048~1.19 μg(r=0.999 8)、0.040~1.00 μg(r=0.999 9)、0.008~0.20 μg(r=0.999 8)、0.080~2.00 μg(r=1.000 0)、0.092~2.31 μg(r=1.000 0)范围内线性关系良好;平均回收率在99.2%~103.9%范围内,RSD范围为0.82%~2.9%。8批漏芦花样品中槲皮素-3-O-α-L-鼠李糖苷、木犀草素、芹菜素、5,7,4'-三羟基-3'-甲氧基黄酮、蜕皮甾酮和泥胡木脂素B葡萄糖苷的含量范围分别为0.66%~1.26%、0.19%~0.60%、0.08%~0.24%、0.03%~0.05%、0.31%~0.99%、0.42%~2.26%。结论:该方法可以用于蒙药漏芦花中6个指标性成分的含量测定,为漏芦花药材的质量控制提供科学依据。

Objective: To establish an HPLC method for simultaneous determination of quercetin-3-O-α-L-rhamnoside,luteolin,apigenin,5,7,4'-trihydroxy-3'-methoxyflavone, ecdysterone and hemislin B glucoside in the flowers of Rhaponticum uniflorum.Methods: The analysis was performed on an YMC-Pack C18 column (4.6 mm×250 mm,5.0 μm)at 35℃,eluted with a gradient program using acetonitrile(A)-0.2% phosphoric acid aqueous solution(B)as the mobile phase.The flow rate was 0.8mL·min-1, and the detection wavelength was 254 nm.Results: The linear ranges of six components in the flowers of R.uniflorum were 0.090-2.24 μg(r=1.000 0)for quercetin-3-O-α-L-rhamnoside, 0.048-1.19 μg(r=0.999 8)for luteolin, 0.040-1.00 μg(r=0.999 9)for apigenin,0.008-0.20 μg(r=0.999 8)for 5,7,4'-trihydroxy-3'-methoxyflavone, 0.080-2.00 μg(r=1.000 0)for ecdysterone,and 0.092-2.31 μg(r=1.000 0)for hemislin B glucoside.The average recoveries of six components were between 99.2%-103.9% with RSD of 0.82%-2.9%.The contents of quercetin-3-O-α-L-rhamnoside, luteolin, apigenin, 5,7,4'-trihydroxy-3'-methoxyflavone, ecdysterone and hemislin B glucoside in eight batches of the flowers of R.uniflorum were 0.66%-1.26%, 0.19%-0.60%, 0.08%-0.24%, 0.03%-0.05%, 0.31%-0.99%, and 0.42%-2.26%.Conclusion: The method established in this research for determination of the six components was feasible and suitable to evaluate the quality of the flowers of R.umuniflorum.
