基于3种黄酮类化合物含量比值鉴别广陈皮道地性

目的:根据不同品种陈皮中3种黄酮类成分橙皮苷、川陈皮素和橘皮素的含量比值,鉴别广陈皮道地性。方法:采用Acquity UPLC BEH C18(2.1 mm×50 mm,1.7 μm)色谱柱,以乙腈-0.1%磷酸水为流动相进行梯度洗脱,流速0.42 mL·min-1,柱温30℃,检测波长283 nm,测定时间12 min;运用UPLC对92批11种不同来源的陈皮药材进行测定,不同品种陈皮中橙皮苷、川陈皮素和橘皮素含量比值不相同,并对其进行系统聚类分析和主成分分析。结果:广陈皮中橙皮苷、川陈皮素和橘皮素含量分别(34.72±1.74)mg·g-1、(3.36±1.62)mg·g-1、(1.83±0.47)mg·g-1,含量比值为19:2:1。系统聚类分析和主成分分析将不同品种陈皮分成4类,能准确把茶枝柑归为一类。其中红橘与茶枝柑的3种黄酮类成分含量比值相近。结论:该法为广陈皮的道地性鉴别提出新方法。

Objective: To identify Citrus reticulata ‘Chachi’ based on the content proportion of hesperidin,nobiletin and tangeretin from various Citri Reticulatae Pericarpium.Methods: An Acquity UPLC BEH C18 column (2.1 mm×50 mm,1.7 μm) was adopted in the study with gradient elution.The mobile phase consisted of acetonitrile and water (with 0.1% phosphoric acid).The flow rate was 0.42 mL·min-1,and the column temperature was maintained at 30℃.The detection wavelength was 283 nm.The system determination time was 12 min.The UPLC method was used for the contents proportion analysis of three flavonoids in 92 samples and 11 kinds of Citri Reticulatae Pericarpium.Cluster analysis and principal component analysis were carried out with different content proportion of hesperidin,nobiletin and tangeretin from various Citri Reticulatae Pericarpium.Results: The contents of hesperidin,nobiletin and tangeretin were (34.72±1.74) mg·g-1,(3.36±1.62) mg·g-1,and(1.83±0.47) mg·g-1,while the proportion of three flavonoids was 19:2:1 in Citrus reticulata ‘Chachi’.Various Citri Reticulatae Pericarpium were divided into 4 clusters and Citrus Reticulata ‘Chachi’ was put to the same branch by cluster analysis and principal component analysis.The contents proportion of three flavonoids in Citrus sinensis and Citrus Reticulata ‘Chachi’ were similar.Conclusion: Our study provides a new method for the identification of Citrus Reticulata ‘Chachi’.
