Objective:To identify donkey hide, the raw material of Asini Colla Corii, and its counterfeits including hide of horse, mule and bovine, based on cytochrome C oxidase subunit 1(COⅠ) barcoding.Methods:The genomic DNA fragments were extracted by optimized methods through universal COⅠ primer and polymerase chain reaction(PCR) condition.The PCR products were bi-directionally sequenced.Sequences were blasted with the sequences from the GenBank.The basic group composition of COⅠ sequence, the inter-and intra-specific variations of COⅠ sequences were analyzed and compared based on Kimura-2-parameter(K2P) distances, and the neighbor-joining method was used to reconstruct phylogenetic tree by Mega 6.0 software to identify donkey hide and its counterfeits.Results:The consistencies of 22 samples were all above 99% by Blast comparison with the data from GenBank.The phylogenetic tree reconstructed by neighbor-joining method could distinguish donkey hide from hide of horse, mule and bovine because of the K2P distance of intra-distance was much larger than that of inter-distance.Conclusion:COⅠ DNA barcoding coupled with PCR offered an efficienct method for origin identification of the raw material of Asini Colla Corii.
