HPLC法同时测定不同产地桔梗中3种聚炔类成分

目的:建立测定桔梗中桔梗炔苷A、桔梗炔苷B和党参炔苷3种聚炔类化合物的HPLC测定方法,并测定我国不同产地桔梗中3种聚炔类化合物的含量。方法:采用加速溶剂萃取法提取,HPLC法测定聚炔类化合物含量。色谱条件:采用依利特Hypersil ODS2(250 mm×4.6 mm,5 μm)色谱柱,以乙腈-水为流动相进行梯度洗脱,流速为1.0 mL·min-1,检测波长为210 nm。结果:不同产地的桔梗药材中聚炔类化合物含量差别较大。在所测的桔梗药材中,山东潍坊的桔梗含桔梗炔苷A最多,为43.3 μg·g-1;湖北十堰产的桔梗中桔梗炔苷B含量最高,为82.3 μg·g-1;四川马尔康的桔梗中党参炔苷含量最高,为461.8 μg·g-1;山东潍坊产的桔梗3种聚炔类化合物总含量最高,为470.3 μg·g-1;3种聚炔类化合物最少的都是吉林延边熏硫处理的桔梗。结论:加速溶剂萃取法提取桔梗中聚炔类化合物效果优于超声法、超临界萃取法和渗漉法,且用时最少,适用于聚炔类化合物提取;建立的HPLC方法操作简便,准确可靠,可用于桔梗药材中3种聚炔类化合物含量的测定。在我国不同产地桔梗药材中,桔梗3种聚炔类化合物含量的差异较大,可作为不同产地桔梗质量控制的一种评价指标。

Objective: To establish an HPLC method for simultaneous determination of platetyolin A, platetyolin B and lobetyolin in Platycodonis Radix. And to determine the contents of these three polyacetylenes in Platycodonis Radix from different habitats. Methods: The contents of three polyacetylenes were determined by accelerated solvent extraction and HPLC quantitative determination. Separation was performed on a Hypersil ODS2 column (250 mm×4.6 mm, 5 μm), with a mobile phase consisting of acetonitrile-water in gradient elution at a flow rate of 1.0 mL·min-1. The detection wavelength was 210 nm. Results: There was a greater difference between the content of polyacetylenes in Platycodonis Radix from different habitats. The highest content of platetyolin A (43.3 μg·g-1), platetyolin B (82.3 μg·g-1), lobetyolin (461.8 μg·g-1), and total content (470.3 μg·g-1) were found in Platycodonis Radix from Weifang Shandong, Shiyan Hubei, Maerkang Sichuan, and Weifang Shandong, respectively. And the lowest polyacetylenes were detected in sulphur fumed sample from Yanbian Jilin. Conclusion: Accelerated solvent extraction is proved to be more effective and less time-consuming than ultrasonic method, supercritical extraction and percolation method, which is suitable for extraction of polyacetylenes. The proposed HPLC method is simple and reliable, which can be used for the content determination of three polyacetylenes in Platycodonis Radix. The contents of polyacetylenes vary greatly in Platycodonis Radix from different habitats, and they can be used as evaluation indexes for quality evaluation of Platycodonis Radix.
