目的:《中华人民共和国药典》(以下简称中国药典)2015年版采用滴定法测定胱氨酸片中胱氨酸的含量,操作烦琐,所以建立简便、快速的氨基酸分析仪测定胱氨酸片中胱氨酸含量的方法。方法:采用直接超声溶解法上机测定,Sykam Cation Separation Coiumn(LCA KO6/Na 4.6 mm×150 mm,7 μm)分离柱,梯度洗脱,分光光度检测器,柱温为57~74℃梯度控温,反应器温度为130℃,检测波长570 nm。结果:胱氨酸在5.01~100.18 μg·mL-1之间呈良好的线性关系(r=0.999 9,n=6),平均加样回收率为100.0%(n=9),检出限为0.26 μg·mL-1,含量测定结果与滴定法相比较,无显著性差异。结论:与中国药典的滴定法比较,该法准确、简易、稳定,适合胱氨酸片中胱氨酸的含量测定。

Objective:The titration method used to measure the content of cystine in cystine tablets,which is applied in Chinese Pharmacopeia 2015,is tedious.In this paper,we meant to establish a simple and rapid amino acid analyzer for the determination of cystine in cystine tablets.Methods:The direct ultrasonic dissolving method was adopted for the determination.Sykam Cation Separation Coiumn (LCA KO6/Na 4.6×150 mm)was used with gradient elution,followed by a spectrophotometric detector. Column temperature between 57-74℃.Thereactor temperature was 130℃.The detection wavelength was 570 nm.Results:Good linearity (r=0.999 9,n=6)can be obtained with the concentration of cystine among 5.01-100.18 μg·mL-1.The average sample recovery rate was 100.0% (n=9)and the detection limit was 0.26 μg·mL-1.Compared with the titration method,there was no significant difference in the detection results. Conclusion:Compared with the titration method in Chinese Pharmacopeia,this method is accurate,simple and stable,which is suitable for the content determination of cystine.
