荧光探针法测定奥沙利铂的方法及机理研究

目的:建立一种简单、快速、灵敏的巴马汀荧光探针法,用于测定抗癌药物奥沙利铂。方法:以奥沙利铂和巴马汀探针对葫芦[7]脲空腔竞争为基础,巴马汀和葫芦[7]脲的水溶液本身无荧光,二者混合后体系荧光显著增强,但当加入无荧光的奥沙利铂后,体系的荧光又显著猝灭。结果:在0.05~1.75 μg·mL-1的范围内奥沙利铂的浓度与荧光猝灭值呈良好的线性关系,最低定量限为20 ng·mL-1,检测限为6 ng·mL-1。并通过核磁共振和密度泛函理论对其作用机理进行了探究,证实奥沙利铂与葫芦[7]脲可以形成1∶1的稳定包合物。结论:该方法可用于血浆中奥沙利铂的测定。

Objective: This paper proposed a simple, rapid and sensitive fluorescence probe method of the palmatine for fluorescence detection of the anticancer drug oxaliplatin.Methods: The methods were based on the competitive reaction of palmatine with oxaliplatin for the occupancy of the cucurbit[7]uril cavity.Oxaliplatin and cucurbit[7]uril are non-fluorescent in aqueous solutions.It was found that the CB[7]-PAL system exhibited high fluorescence in aqueous solution, which was quenched remarkably in the presence of L-OHP.Results: The fluorescence quenching values(ΔF) showed good linear relationship with L-OHP concentrations from 0.05 μg mL-1 to 1.75 μg mL-1 with a quantitative detection limit of 20 ng·mL-1 and a detection limit of 6 ng·mL-1.The mechanism was studied by 1H NMR and density functional theory, which confirmed that oxaliplatin and cucurbit[7]uril can form a 1:1 stable inclusion complex.Conclusion: The proposed method was applied successfully to the determination of oxaliplatin in plasma.
