RP-HPLC同时测定地黄饮子中松果菊苷、毛蕊花糖苷、马钱苷的含量

目的:建立反相高效液相色谱法(RP-HPLC法)同时测定地黄饮子中松果菊苷、毛蕊花糖苷、马钱苷的含量。方法:采用Kromasil C18(150 mm×4.6 mm,5 μm)色谱柱,以甲醇-0.01%磷酸水溶液为流动相进行梯度洗脱,流速1.0 mL·min-1,检测波长为334 nm(松果菊苷及毛蕊花糖苷)和240 nm(马钱苷),进样量20 μL。结果:松果菊苷、毛蕊花糖苷、马钱苷线性范围分别为3.37~67.40 μg·mL-1r=0.999 2)、3.31~66.20 μg·mL-1r=0.999 3)和5.76~115.2 μg·mL-1r=0.999 6),平均加样回收率(n=6)分别为97.2%、98.1%和98.5%。6批样品中松果菊苷、毛蕊花糖苷、马钱苷含量分别为73.25~81.36、51.54~60.28、14.77~17.08 μg·mL-1结论:本方法可用于地黄饮子中松果菊苷、毛蕊花糖苷及马钱苷的含量测定。

Objective: To establish an RP-HPLC method for simultaneous determination of echinacoside,verbascoside and loganin in Dihuangyinzi. Methods: A Kromasil C18 column(150 mm×4.6 mm,5 μm)was adopted as analytical column. The mobile phase consisted of methanol-0.01% phosphoric acid with gradient elution at a flow rate of 1.0 mL·min-1,the detection wavelengths were 334 nm for echinacoside and verbascoside,240 nm for loganin,and the injection volume was 20 μL. Results: The linear response ranges observed were 3.37-67.40 μg·mL-1(r=0.999 2),3.31-66.20 μg·mL-1(r=0.999 3)and 5.76-115.2 μg·mL-1(r=0.999 6),respectively;the average recoveries(n=6)were 97.2%,98.1% and 98.5%,respectively. The contents of echinacoside,verbascoside,loganin in six batches of samples were 73.25-81.36,51.54-60.28 and 14.77-17.08 μg·mL-1,respectively. Conclusion: The method can be used for simultaneous determination of sechinacoside,verbascoside and loganin in Dihuangyinzi.
