茅苍术麸炒前后对乙酸致胃溃疡大鼠胃动素和胃泌素的影响

目的: 考察茅苍术麸炒后对乙酸致胃溃疡大鼠血清及胃组织中胃动素(MTL)和胃泌素(GAS)的影响,初步探讨茅苍术麸炒后抗胃溃疡作用变化的机理。方法: 实验随机分成10 组,即假手术组、模型组、奥美拉唑组、三九胃泰组、茅苍术生品低剂量组、茅苍术生品中剂量组、茅苍术生品高剂量组,茅苍术麸炒品低剂量组、茅苍术麸炒品中剂量组和茅苍术麸炒品高剂量组。参考Okabe 方法进行适当改良,即外科手术法以乙酸诱导大鼠胃溃疡模型。采用常规组织病理学HE 染色法观察胃组织病理改变情况,采用酶联免疫吸附(ELISA)法检测血清及胃组织中MTL 和GAS 的含量。结果: 茅苍术麸炒品抗胃溃疡的作用优于生品。与假手术组比较,模型组大鼠血清及胃组织中MTL 和GAS 的含量明显降低;与模型组比较,茅苍术生品和麸炒品各组血清及胃组织中MTL 和GAS 的含量均有不同程度的升高,麸炒品的作用优于生品。结论: 茅苍术麸炒后抗胃溃疡的作用增强,其作用机理部分是由于促进MTL 和GAS 的分泌。

Objective: To investigate the effect of Rhizoma Atractylodis Lanceae on levels of motilin(MTL) and gastrin(GAS)in the rat model of gastric ulcer induced by acetic acid after processing,and evaluate the mechanisms of action involved in the anti-ulcer properties of Rhizoma Atractylodis Lanceae. Methods: All rats were randomly divided into the following 10 groups:sham-operated group,model group,omeprazole group, Sanjiu Weitai granule,low dose group of the crude Rhizoma Atractylodis Lanceae,middle dose group of the crude Rhizoma Atractylodis Lanceae,high dose group of the crude Rhizoma Atractylodis Lanceae,low dose group of the processed Rhizoma Atractylodis Lanceae,middle dose group of the processed Rhizoma Atractylodis Lanceae,and high dose group of the processed Rhizoma Atractylodis Lanceae. The model of gastric ulcer was established by acetic acid in rats according to the method of Okabe et al. with some modifcations. Histopathological changes were evaluated by hematoxylin and eosin(HE)stain. The levels of MTL and GAS in serum and gastric tissues were determined by the double-antibody sandwich enzyme-linked immunosorbent assay(ELISA). Results: The results demonstrated that the processed Rhizoma Atractylodis Lanceae had better anti-ulcer effect compared with the crude Rhizoma Atractylodis Lanceae. The productions of MTL and GAS in serum and stomachs of model group were notably decreased compared with sham-operated group.Compared with model group,higher levels of MTL and GAS in serum and stomachs of the crude Rhizoma Atractylodis Lanceae and Rhizoma Atractylodis Lanceae processed groups were observed. Compared with the crude Rhizoma Atractylodis Lanceae,the processed Rhizoma Atractylodis Lanceae was more effective. Conclusion: The results show that the anti-ulcer effect of Rhizoma Atractylodis Lanceae is enhanced after processing. The anti-ulcer effect of Rhizoma Atractylodis Lanceae can be partly attributed to up-regulation of MTL and GAS.
