凌霄花HPLC-ELSD指纹图谱的研究

目的:采用HPLC-ELSD法构建凌霄花药材的指纹图谱,并对其主要成分进行鉴定。方法:采用Thermo C18(4.6 mm×250 mm,5μm)色谱柱,以甲醇(A)-0.1%甲酸水溶液(B)为流动相,梯度洗脱,流速0.8 mL·min-1,柱温30℃,ELSD漂移管温度105℃,氮气流速3.5 L·min-1。结果:对不同产地的10批凌霄花药材进行分析,建立了凌霄花药材HPLC-ELSD指纹图谱,标定共有峰12个。通过对照品指认及HPLC-TQ-MS进一步分析,鉴定出了其中6个色谱峰,分别为麦角甾苷、顺式麦角甾苷或异麦角甾苷、芹菜素、科罗索酸、齐墩果酸和熊果酸。结论:该方法准确可靠、重复性好,可用于凌霄花药材的整体质量评价。

Objective: To establish the fingerprints of Campsis grandiflora(Thunb.) K.Schum.by HPLC-ELSD and elucidate the main components.Methods: Separation was performed on a Thermo C18(4.6 mm×250 mm, 5 μm) column.Gradient elution was performed by mobile phase consisting of methanol and 0.1% formic acidwater with the flow rate of 0.8 mL·min-1.Column temperature and ELSD drift tube temperature were controlled at 30℃ and 105℃, respectively.The nitrogen flow rate was 3.5 L·min-1.Results: Totally 10 batches of Campsis grandiflora(Thunb.) K.Schum.crude drugs from different regions were analyzed.Perfect fingerprints were obtained by HPLC-ELSD which can be used for evaluation of Campsis grandiflora(Thunb.) K.Schum.12 common peaks were confirmed in fingerprints.6 compounds were elucidated by reference standards and HPLC-TQ-MS, including acteoside, cisacteoside or isoacteoside, apigenin, corosolic acid, oleanolic acid and ursolic acid.Conclusion: The HPLC-ELSD characteristic fingerprint of Campsis grandiflora(Thunb.) K.Schum. is accuracte and reproducible, and the established method can be applied to the quality assessment of Campsis grandiflora(Thunb.) K.Schum.
