目的:建立HPLC法同时测定森登-4有效部位中7种成分的含量。方法:采用Extend-C18(250mm×4.6 mm,5μm)色谱柱,以乙腈(A)-0.6%磷酸水溶液(B)为流动相,梯度洗脱,流速0.5 mL·min-1,柱温27℃,检测波长为254 nm和292 nm,进样量10μL。结果:没食子酸、京尼平-1-β-D-龙胆二糖苷、栀子苷、(2R,3R)-双氢杨梅素、芦丁、杨梅素、槲皮素的质量浓度与其峰面积具有良好的线性关系(r=0.9999);平均回收率为97.30%~103.9%,RSD为0.08%~1.00%。没食子酸、京尼平-1-β-D-龙胆二糖苷、栀子苷、(2R,3R)-双氢杨梅素、芦丁、杨梅素、槲皮素平均含量分别为0.379%、0.447%、2.740%、3.965%、0.253%、0.878%、0.044%。结论:经方法学验证,本法可用于森登-4有效部位的化学成分含量测定。

Objective: To establish an HPLC method for simultaneous determination of the content of 7 effective components in Mongolian medicine Sendeng-4.Methods: The Extend-C18 column(250 mm×4.6 mm, 5 μm) was adopted with a mobile phase consisting of acetonitrile(A) and 0.6% phosphoric acid aqueous solution(B) with gradient elution at the flow rate of 0.5 mL·min-1.The column temperature was 27℃, the detection wavelengths were 254 nm and 292 nm, and the injection volume was 10 μL.Results: Mass concentration and the peak area of gallic acid, genipin-1-β-D-gentiobioside, geniposide, (2R, 3R)-dihydromyricetin, rutin, myricetin and quercetin exhibited a good linear relationship with correlation coefficient of 0.999 9, the average recovery was 97.30%-103.94% and the RSD was 0.08%-1.00%.The average contents of gallic acid, genipin-1-β-Dgentiobioside, geniposide, (2R, 3R)-dihydromyricetin, rutin, myricetin, quercetin of 6 parts test solution were 0. 379%, 0.447%, 2.740%, 3.965%, 0.253%, 0.878%, and 0.044%, respectively.Conclusion: The results of methodology validation showed that the established method can be applied to the determination of chemical constituent of Mongolian medicine Sendeng-4.
