瑶药旱田草的定性定量定法研究

目的:建立旱田草药材的定性定量方法,考察广西不同产地、不同采收时节的药材质量。方法:采用TLC法对旱田草进行定性鉴别,展开剂为乙酸丁酯-甲醇-甲酸-水(6:1.5:1.5:1);采用HPLC法对旱田草中的毛蕊花糖苷进行含量测定,使用Waters SunFire C18色谱柱(4.6 mm × 250 mm,5 μm),柱温35 ℃,以甲醇-0.4%磷酸溶液(32:68)为流动相,流速1.0 mL·min-1,检测波长334 nm;参照中国药典2010年版一部测定水分、灰分和浸出物。结果:薄层色谱中斑点清晰,分离度好;毛蕊花糖苷质量浓度在2.002~40.04 μg·mL-1(r=1.000 0)范围内线性关系良好,加样回收率为98.61%(RSD=1.7%,n=6);所测的12批样品含量在0.196~16.76 mg·g-1之间。结论:本文建立的方法可用于瑶药旱田草的质量控制。

Objective:To establish a qualitative and quantitative method for Lindernia ruellioides (Colsm.)Pennell,and investigate the quality of this medicine from different habitats and harvest seasons in Guangxi.Methods:The TLC method was used for qualitative identification and butyl acetate-methanol-formic acid-water(6:1.5:1.5:1)was used as the developing agent;and the determination of verbascoside was conducted by HPLC using a column of Waters SunFire C18(4.6 mm×250 mm,5 μm),the mobile phase was methanol-0.4% phosphoric acid(32:68)at a flow rate of 1.0 mL·min-1;the detection wavelength was 334 nm,and the column temperature was 35 ℃.The contents of moisture,total ash and ethanol-soluble extractives were determined with the methods in Chinese Pharmacopeia(ChP 2010).Results:The spot of verbascoside could be well identified by TLC.In the system of HPLC,verbascoside showed good linear relationship in the ranges of 2.002-40.04 μg ·mL-1(r=1.000 0),The average recovery was 98.61%(RSD=1.7%,n=6).The contents in 12 batches of samples were between 0.196 mg·g-1 to 16.76 mg·g-1.Conclusion:The qualitative and quantitative method established can be used for the quality control of Lindernia ruellioides (Colsm.)Pennell.
