目的:建立同时测定黄连上清片中"黄芩-黄连-黄柏"药对中黄柏碱、黄柏酮、小檗碱、巴马汀、黄芩苷、黄芩素、汉黄芩素、吴茱萸内酯和阿魏酸含量的HPLC分析方法。方法:采用Boston Green ODS色谱柱(4.6 mm×250 mm,5μm),流动相为乙腈-0.1%磷酸水溶液(含0.02 mol·L-1磷酸二氢钠),梯度洗脱,柱温35℃,检测波长为210 nm。结果:本实验方法学考察结果表明各色谱峰间分离度均达到定量要求,精密度、回收率、重复性等参数均达到定量分析的要求。12批次样品的含量结果显示9个成分中有7个被检测出,黄柏酮和阿魏酸未检出。和单味中药材成分相比,推断考虑在处方工艺制备过程中存在成分损失。结论:本实验针对黄芩-黄连-黄柏药对,建立了同时测定黄连上清片中10个指标性成分的含量分析方法,简便可靠,可用于黄连上清片的质量控制。

Objective: To established an HPLC method for simultaneous determination of 9 components (as phellodendrine, obacunone, berberine, palmatine, baicalin, baicalein, wogonin, evodine and ferulic acid)in couplet medicines Scutellariae Radix-Coptidis Rhizoma-Phellodendri Chinensis Cortex. Methods:The Boston Green ODS column (4.6 mm×250 mm,5 μm) was used, and the mobile phase was acetonitrile-0.1% phosphoric acid solution for gradient elution. The column temperature was 35℃, and the detection wavelength was 210 nm. Results: Methodology results showed that separation of peaks, precision, recovery, repeatability and so on met the requirements. Totally 7 in 9 components were detected out in 12 batches of samples by this method. Obacunone and ferulic acid were not detected out. The possible cause was the content loss in processing procedures. Conclusion: Targeting for couplet medicine Scutellariae Radix-Coptidis Rhizoma-Phellodendri Chinensis Cortex, the analytical method established in this paper for simultaneous determination of 10 index components in Huanglian Shangqing tablets was simple and accurate for quality control of the modicine.
