炮制前后淫羊藿不同提取物对ACE、α-葡萄糖苷酶的抑制活性及对亚硝酸根的清除活性研究

目的: 考察淫羊藿炮制前后的不同提取物对血管紧张素转化酶(ACE)、α-葡萄糖苷酶抑制活性及亚硝酸根清除活性的变化。方法: 采用高效液相色谱法比较淫羊藿炮制前后的不同提取物对ACE的抑制作用,采用分光光度法测定其对亚硝酸盐的清除率以及对α-糖苷酶的抑制活性。结果: 对ACE的抑制活性,只有石油醚提取物表现出了活性,并且未经炮制的淫羊藿明显强于炮制后的。对α-糖苷酶的抑制活性,未经炮制的淫羊藿二氯甲烷、乙酸乙酯、甲醇、水提取物及其水煎液均有活性;炮制后淫羊藿的二氯甲烷提取物则无活性,且其他3个提取物和水煎液的活性降低,但不明显。对亚硝酸根的清除活性,炮制后的淫羊藿石油醚、二氯甲烷、乙酸乙酯提取物和水煎液均表现一定的活性,未经炮制的淫羊藿的活性主要体现在二氯甲烷、乙酸乙酯、甲醇提取物和水煎液,其中水煎液比任何一个提取物的活性都好。本文也对炮制前后淫羊藿不同提取物、水煎液中淫羊藿苷的含量做了对比,结果显示淫羊藿苷可以在乙酸乙酯、甲醇、水提取物和水煎液中被检测到,炮制后淫羊藿的各个提取物的淫羊藿苷的含量均低于未炮制的。结论: 淫羊藿经炮制后,其有效成分淫羊藿苷含量有所降低,且其对ACE、α-葡萄糖苷酶及亚硝酸根的抑制活性均有所降低。

Objective: To determine the activity of angiotensin converting enzyme (ACE) and α-glucosidase inhibition, and the nitrite scavenging from different extracts and water decoction of unprocessed and processed Epimedium brevicornum Maxim.. Methods: The unprocessed and processed E.brevicornum were extracted with different solvent, and the ACE inhibitory activity was determined by HPLC.The extracts' nitrite scavenging capability and their α-glucosidase inhibitory activity were measured by spectrophotometry, respectively. Results: For the ACE inhibitory activity analysis, the results showed that it only appears in petroleum ether extract, and the unprocessed IC50 was lower than the processed samples.For α-glycosidase inhibitory activity analysis, the activity was shown in dichloromethane, ethyl acetate, methanol, water extracts and water decoction of the unprocessed E.brevicornum, but the dichloromethane extract of the processed exhibited no inhibitory activity.In addition, the inhibitory activity of the other extracts and water decoction of the processed was also lower than the unprocessed, but no significant differences were observed.In the aspect of nitrite scavenging activity, the processed E.brevicornum showed a certain activity in petroleum ether, dichloromethane, ethyl acetate extracts and water decoction, while the unprocessedshowed a certain ability of scavenging activity in dichloromethane, ethyl acetate, the methanol extracts and water decoction.The icariin's contents of different extracts and water decoction of unprocessed and processed E.brevicornum were determined by HPLC.The results showed that icariin can be detected in ethyl acetate, methanol, water extracts and water decoction, meanwhile, the unprocessed E.brevicornum generally show a higher content than the processed. Conclusion: For, The extracts of unprocessed E.brevicornum show a higher activity of ACE and α-glucosidase inhibition, and nitrite scavenging than that of the processed.
