一测多评法同时测定五味子中8个木脂素类成分的含量

目的: 采用"一测多评"法同时测定五味子中8个木脂素类(五味子醇甲、戈米辛J、五味子醇乙、戈米辛G、五味子酯甲、五味子甲素、五味子乙素和五味子丙素)有效成分的含量. 方法: 以五味子为研究对象,以五味子醇甲为内参物,建立五味子醇甲与其他7个成分相对校正因子;采用外标法测定五味子中五味子醇甲的含量,通过相对校正因子计算其他7个成分的含量,并将该方法的测定结果与外标法测定的结果比较. 结果: 五味子醇甲、戈米辛J、五味子醇乙、戈米辛G、五味子酯甲、五味子甲素、五味子乙素和五味子丙素进样量分别在0.205~4.104 μg(r=0.999 5)、 0.015 8~0.317 μg(r=1.000)、0.060 9~1.218 μg(r=1.000)、0.011 7~0.234 μg(r=1.000)、0.010 4~0.208 μg(r=1.000)、0.059 4~1.188 μg(r=1.000)、0.094 9~1.898 μg(r=1.000)、0.010 0~0.200 0 μg(r=0.999 9)线性关系良好,平均加样回收率(n=9)分别为100.5%、98.9%、100.3%、101.2%、101.3%、102.0%、100.4%、102.6%.在一定线性范围内,五味子醇甲与戈米辛J、五味子醇乙、戈米辛G、五味子酯甲、五味子甲素、五味子乙素和五味子丙素间的相对校正因子分别为1.130 8、1.033 2、0.856 4、0.850 5、1.042 0、1.086 4、1.032 8,且在不同实验条件下重现性良好[相对校正因子的RSD(n=6)< 2.5%],12批五味子的"一测多评"法计算值和外标法测定值间无显著差异,实验所得的校正因子可信. 结论: 在对照品缺乏的情况下,以外标法测定五味子醇甲,利用相对校正因子实现对其他7个木脂素成分含量测定可行.

Objective: To establish a quantitative analysis method for determining contents of eight lignans components(schisandrin,gomisin J,schisandrol B,gomisin G,schisantherin A,deoxyschizandrin,schizandrin B,and schizandrin C)from the fruits of Schisandra chinensis by using multi-components with a single-marker(QAMS). Methods: With the typical composition schisandrin as the indexes,the relative correction factors(RCFs)between the schisandrin and the other seven lignans components were established to detect the quantitation of schisandrin,and the contents of these two ingredients in all samples were also determined using the external standard method. Results: Good linear relationships were observed for schisandrin,gomisin J,schisandrol B,gomisin G,schisantherin A,deoxyschizandrin,schizandrin B,and schizandrin C within the ranges of 0.205-4.104 μg(r=0.999 5),0.015 8-0.317 μg(r=1.000 0),0.060 9-1.218 μg(r=1.000),0.011 7-0.234 μg(r=1.000),0.010 4-0.208 μg(r=1.000),0.059 4-1.188 μg(r =1.000),0.094 9-1.898 μg(r=1.000)and 0.010 0-0.200 0 μg(r =0.999 9),respectively.The average extraction recoveries(n=9)were 100.5% for schisandrin,98.9% for gomisin J,100.3% for schisandrol B,101.2% for gomisin G,101.3% for schisantherin A,102.0% for deoxyschizandrin,100.4% for schizandrin B,and 102.6% for schizandrin C,respectively.The RCFs of gomisin J,schisandrol B,gomisin G,schisantherin A,deoxyschizandrin,schizandrin B,and schizandrin C with reference to schisandrin were 1.130 8,1.033 2,0.856 4,0.850 5,1.042 0,1.086 4 and 1.032 8,and the repeatability was good under different experimental conditions[RSDs of RCFs(n=6) <2.5%].There were no significant differences between the calculated value and estimated value on QAMS and external standard method of the eight lignans in 12 batches of the fruits of Schisandra chinensis.In addition,the results showed that the RCF was authentic. Conclusion: In the case of lacking reference substance,the quantitation of schisandrin could be detected.It is feasible to take advantage of the RCF to quantitate eight lignans components.
