左金方、黄连、吴茱萸诱导人胃癌细胞SGC-7901凋亡的作用比较

目的: 比较左金方、黄连、吴茱萸对人胃癌细胞SGC-7901生长的活性抑制和凋亡诱导作用. 方法: 采用MTT比色法观察左金方、黄连、吴茱萸对SGC-7901的生长抑制情况;流式细胞仪PI单染亚二倍体峰分析细胞凋亡率;Hoechst 33258染色观察细胞形态学变化以及Western blot方法检测左金方、黄连、吴茱萸对人胃癌细胞SGC-7901中Bcl-2、Bax蛋白表达的影响. 结果: 左金方和黄连分别作用SGC-7901细胞后,均能抑制细胞活性,且与时间呈依赖关系;1.0 mg·mL-1左金方和对应浓度的黄连(0.857 mg·mL-1)作用于SGC-7901细胞48 h,细胞凋亡率分别为(36.40±1.59)%和(21.90±0.27)%,与对照组(1.69±1.91)%比较差异均有统计学意义(P <0.01),组间比较差异均有统计学意义(P <0.01),对应浓度的吴茱萸(0.143 mg·mL-1)几无作用;Hochest染色细胞出现核固缩状和颗粒状荧光等典型的凋亡学特征;左金方和黄连均可降低SGC-7901细胞Bcl-2基因表达和增加Bax基因表达. 结论: 左金方与黄连均可通过上调Bax/Bcl-2的比率诱导胃癌细胞的凋亡,但左金方的作用优于黄连(P <0.01),吴茱萸几无作用.

Objective: To compare the cellular growth inhibition and apoptosis of human gastric carcinoma cells SGC-7901 induced by traditional decoction of Zuojinfang,Coptidis Rhizoma and Evodiae Fructus. Methods: The SGC-7901 were treated with different traditional decoction of Zuojinfang,Coptidis Rhizoma and Evodiae Fructus,and the cellular growth inhibition was measured by MTT;the apoptotic rate was evaluated by FCM and the morphology was observed by Hoechst 33258 staining. Detection of Bcl-2,Bax gene and protein expression was carried out by using Western blot method. Results: After the cells treated by traditional decoction of Zuojinfang,Coptidis Rhizoma,the cell survival rates decreased in a time-dependent manner. When the cells were treated with 1.0 mg·mL-1 traditional decoction of Zuojinfang and 0.857 mg·mL-1 Coptidis Rhizoma for 48 h,the percentages of apoptosis were (36.40±1.59)% and (21.90±0.27)%,respectively.The differences in comparison with the control group (1.69±1.91)% were statistically significant (P <0.01),and the differences in comparison between the traditional decoction of Zuojinfang group and Coptidis Rhizoma group were also statistically significant (P <0.01). No effect was observed at the same concentrations of Evodiae Fructus (0.143 mg·mL-1). Hochest staining results showed cells nuclear condensation,granular fluorescence and other typical features of apoptosis. Both of the traditional decoctions of Zuojinfang and Coptidis Rhizoma could reduce the Bcl-2 gene expression and increase Bax gene expression. Conclusion: Both of the traditional decoctions of Zuojinfang and Coptidis Rhizoma could significantly inhibit the growth of SGC-7901 and induce the cell apoptosis via increasing the ratio of Bax/Bcl-2,where.The effect of traditional decoction of Zuojinfang was stronger than that of Coptidis Rhizoma under the same conditions,and Evodiae Fructus almost had no effect.
