薄层色谱扫描法对减肥保健食品中酚酞的定性定量分析

目的:建立基层适宜的减肥保健食品中添加酚酞的分析方法。 方法:采用薄层色谱扫描(TLCS)法。供试品用无水乙醇超声提取,以GF254铝合金硅胶板为固定相,以乙酸乙酯-石油醚(60~90 ℃)-甲醇(10∶6∶1)为展开剂,采用单波长吸收扫描法,λ=254 nm,狭缝宽8 mm×0.6 mm。 结果:酚酞标准曲线线性范围为4.00~20.0 μg(r=0.996),平均回收率(n=6)为100.2%。用建立的TLCS法分别对减肥保健食品中共计16个品种20个批次进行酚酞的检测,11个批次检出了酚酞,含量在1.37%~16.51%。 结论:所建立的方法经与高效液相色谱-质谱(HPLC-MS)方法比较,可作为减肥保健类产品添加酚酞的定性和定量分析方法。

Objective: To establish a suitable analysis method for identification and determination of phenolphthalein added into anti-obesity and healthcare food. Methods: Using TLCS method,the sample was extracted with anhydrous alcohol using ultrasonic extraction.The stationary phase was the GF254aluminium alloy silica gel plate while the developing solvent was a mixture of acetic ether,petroleum ether(60~90 ℃)and methanol(10∶ 6∶ 1).TLCS single wave-length absorption was used,λ=254 nm,narrow slot:8 mm×0.6 mm. Results: The calibration curve was linear in the range of 4.00-20.0 μg(r=0.996)for phenolphthalein,and the average recovery(n=6)was 100.2% for phenolphthalein.The established TLCS method was used to test a total of 16 varieties(20 batches)of anti-obesity and healthcare food,among which 11 healthcare products were detected with phenolphthalein,the content of which ranged 1.37%-16.51%. Conclusion :The established method in comparison with HPLC-MS can be used as the identification and determination method of phenolphthalein added into the products.
