目的 : 利用HPLC-UV特征图谱分析方法,建立山楂药材的特征图谱。方法:采用聚酰胺色谱柱对样品进行净化处理;利用HPLC-UV法测定山楂药材特征图谱,采用Ultimatetm AQ-C18(4.6 mm×250 mm,5 μm)色谱柱,以乙腈(A)-0.25%磷酸水溶液(B)为流动相,梯度洗脱(0 min,12%A;22 min,15%A;30 min,16%A;65 min,20%A;90 min,40%A),流速1.0 mL·min-1,检测波长360 nm,柱温30 ℃。 结果:10批药材的相似度在0.95以上,建立了山楂药材的HPLC-UV特征图谱,确定了10个共有峰,并用对照品确认了其中的4个共有峰(绿原酸、芦丁、金丝桃苷和异槲皮苷)。 结论:该方法操作简便,重现性好,可为山楂质量控制方法提供参考。

Objective: To establish a method of characteristic chromatogram analysis on hawthorn berries by HPLC-UV. Methods :The sample was pre-treated by polyamide solid phase extraction column.The gradient elution mode(0 min,12%A;22 min,15%A;30 min,16%A;65 min,20%A;90 min,40%A) was applied in chromatographic separation.A UltimateR AQ-C18 (4.6 mm×250 mm,5 μm) column was used with the mobile phase of acetonitrile(A)-water(0.25%phosphoric acid)(B),the flow rate of 1.0 mL·min-1,the UV detection wavelength of 360 nm. Results: The similarities of 10 batches of samples were over 0.95. The characteristic chromatogram of hawthorn berries was established and 10 common peaks were found.Among them,4 peaks(chlorogenic acid,rutin,hyperoside and isoquercitrin) were identified by comparison with the references. Conclusion: The established method shows satisfactory accuracy,stability,and reproducibility,and can be used as a quality control method for hawthorn berries.
