Objective: To establish a method for simultaneous determination of chlorogenic acid,geniposide,berberine,baicalin,aloe-emodin,rhein,emodin,chrysophanol and physcion in Zhizi Jinhua pills. Methods: The separation was performed on a DIKMA Platisil ODS column(250 mm×4.6 mm,5 μm)with gradient elution(0-5 min,40%A;5-20 min,40%A→60%A;20-26 min,60%A→60%A;26-35 min,60%A→90%A;35-52 min,90%A;52-54 min,90%A→40%A)using methanol and 0.1% phosphoric acid aqueous solution as the mobile phase.The flow rate was 1 mL·min-1 and the detection wavelengths were 327 nm for chlorogenic acid and berberine,254 nm for geniposide,baicalin,aloe-emodin,rhein,emodin,chrysophanol and physcion,respectively. Results: Linear ranges of chlorogenic acid,geniposide,berberine,baicalin,aloe-emodin,rhein,emodin,chrysophanol and physcion concentrations were 0.02-0.4 μg(r=0.9998),0.049-0.98 μg(r=1.000),0.05-1 μg(r=0.9999),0.1-2 μg(r=1.000),0.0025-0.05 μg(r=0.9996),0.001-0.02 μg(r=0.9997),0.0014-0.028 μg(r=0.9998),0.004-0.08 μg(r=0.9998),0.00033-0.0066 μg(r=0.9998),respectively.Their average recoveries fell between 98.8% and 100.4%. Conclusion: The methodology validation shows that this HPLC method established can be used to determine the contents of chlorogenic acid,geniposide,berberine,baicalin,aloe-emodin,rhein,emodin,chrysophanol and physcion in Zhizi Jinhua pills.The method is an effective method for the quality control of Zhizi Jinhua pills.
