目的:建立一种肺癌组织中微量元素亚细胞分布的分析方法。方法:采用电感耦合等离子体质谱法(ICP-MS)研究了肺癌组织、癌旁组织和正常肺组织中Cr、Mn、Fe、Co、Cu、Zn、Se、Cd 8个微量元素在细胞核、线粒体、溶酶体、微粒体和细胞上清液等亚细胞组分中的分布。结果:除Se元素外,其余各元素在亚细胞组分中均呈非均匀分布,采用t检验进行统计比较,所有待测元素在肺癌组织、癌旁组织和正常肺组织的亚细胞分布中均存在不同程度的显著性差异,在细胞核、线粒体、溶酶体、微粒体、细胞上清液等5个亚细胞组分中,肺癌组织中Cr、Mn、Fe、Co、Zn、Cd 6个元素的含量均低于癌旁组织和正常肺组织,而Se和Cu的含量高于癌旁组织和正常肺组织。结论:采用ICP-MS法可实现肺癌组织中微量元素亚细胞分布的准确分析,微量元素的变化与肺癌的发生、发展密切相关,对肺癌过程变化的研究和指导临床治疗均具有重要意义。

Objective: To establish a new method for determination of subcellular distribution of trace elements in lung cancer tissue. Methods: Cr,Mn,Fe,Co,Cu,Zn,Se and Cd elements were determined in the subcellular fractions of nuclei,mitochondria,lysosome,microsome and cytosol of the lung cancer tissue,the para-carcinoma tissue and the normal lung tissue by inductively coupled plasma mass spectrometry (ICP-MS). Results: The analysis result of the samples showed that except for Se,the other elements were heterogeneously distributed in the subcellular component.With t test,the statistical comparison showed that all the analytic elements in the subcellular distribution of the lung cancer tissue,the para-carcinoma tissue and the normal lung tissue were significantly different in varying degrees.In the five subcellular components such as cell nucleus,mitochondria,lysosome,microsome and cell supernatant,the contents of six elements like Cr,Mn,Fe,Co,Zn and Cd in the lung cancer tissue were all lower than the ones in the para-carcinoma tissue and the normal lung tissue while the contents of Se and Cu were higher than the ones in the para-carcinoma tissue and the normal lung tissue. Conclusion: ICP-MS is a useful tool for simultaneous determination of multi-elements in the subcellular fractions of lung cancer tissue,the para-carcinoma tissue and the normal lung tissue.The result indicates that the change of concentration of trace elements is closely related to the generation and development of lung cancer,and the change of the trace elements are related to occurrence and development of lung cancer.
