2种不同设计方法优化红梅消总皂苷双水相提取工艺研究

目的:确定红梅消中总皂苷的最佳提取工艺。 方法:以红梅消总皂苷含量为指标,通过星点设计与混料设计2种不同设计方法优化双水相提取中各相质量百分比,预测出双水相提取红梅消总皂苷的最优工艺。 结果:预测值与实测值无显著差异。星点设计法总皂苷的最佳提取条件:硫酸铵12.14%、乙醇40.01%、总皂苷的含量为2.95%;混料设计法总皂苷的最佳提取条件:硫酸铵11%,乙醇37%、总皂苷的含量为3.10%。 结论:2种设计方法均适用于优化双水相提取中各相质量百分比,可行性高,结果可靠;双水相体系对红梅消总皂苷有较好的提取作用,工艺简单、节能、易操作,且易放大于工业生产。

Objective: To obtain the optimal extraction of saponins from Rubus parvifolius L.. Methods: Using the total content of saponins from Rubusparvifolius L. as an index,the modified central composite design and the mixture design were used to optimize the weight percents of different phases in the aqueous two-phase system,and to predict the optimal process conditions for aqueous two-phase extraction of saponins from Rubus parvifolius L.. Results: There was no significant difference between the predicted value and the measured value.The optimal process conditions by the central composite design were as follows:12.14% of ammonium sulfate and 40.01% of ethyl alcohol.The extraction yield of saponin sunder this conditionswas 2.95%.And the optimal process conditions by the mixture design were as follows:11%of ammonium sulfate and 37% of ethyl alcohol.Under this condition,the yield was 3.10%. Conclusion: Both designs are applicable for optimizing the weight percentages of different phases in the aqueous two-phase system,with good practicability and high reliability.The aqueous two-phase system is an effective method for extracting the saponins from Rubus parvifolius L.The method is simple,energy-efficient,convenient and readily enlargable.
