目的: 以不同产地的甘遂为研究对象,建立了甘遂的HPLC 指纹图谱及指标成分大戟二烯醇的含量测定方法,为科学评价和有效控制甘遂药材的质量提供科学依据。 方法: 采用Agilent Eclipse XBD-C8色谱柱(150 mm×4.6 mm,5 μm);以水与乙腈为流动相,梯度洗脱,流速1.0 mL·min-1,检测波长210 nm,柱温40℃,进样量10 μL。 结果: 10 批次甘遂药材中,有9 个共有峰,经相似度计算,整体相似性好,并同步测定了甘遂中大戟二烯醇的含量。 结论: 本文建立的甘遂指纹图谱及大戟二烯醇含量测定的方法有较强的针对性,为甘遂药材的研究以及制定甘遂药材的质量控制标准提供科学依据。

Objective: To establish fingerprint and quantitative analysis of Kansui Radix originated from different areas by HPLC,and provide reasonable method for quality evaluation and quality control of crude materials. Methods: Euphol was used as the standard substance to conduct quantitative analysis and to establish the HPLC fingerprint for Kansui Radix by HPLC.The Agilent Eclipse XBD-C8 column (150 mm×4.6 mm,5 μm) was used and the wavelength of UV detection was 210 nm.The column temperature was 40℃.The mobile phases were acetonitrile and water eluting in a linear gradient mode at the flow rate of 1.0 mL·min-1. Results: Nine common peaks were selected as markers in HPLC fingerprint of ten batches of samples of Kansui Radix,and the overall similarity was good according to the similarity calculation.The content of euphol was also determined simultaneously. Conclusion: Fingerprint and quantitative analysis of Kansui Radix was satisfactory and specific in this study.Methods established could be used for research and establishment of quality control standard for Kansui Radix in future.
