6-羟多巴胺损伤致急性帕金森氏病大鼠模型的对比分析

目的: 应用高效液相色谱-电化学法(HPLC-ECD)测定大鼠脑微透析液中单胺类神经递质及其代谢产物,检测2种帕金森氏病(PD)大鼠模型的成功率。 方法: 制作在右侧同一位点不同深度及不同位点同一深度注射6-羟多巴胺(6-OHDA)-帕金森氏病大鼠模型,术后7 d将探针插入大鼠右侧纹状体,在清醒自由活动状态下,用Ringer氏液以1.5 μL·min-1的速度灌流,每20 min收集1管透析液,将其注入HPLC-ECD,并对其中所含的多巴胺(DA)、3,4-二羟苯乙酸(DOPAC)、高香草酸(HVA)进行含量测定。 结果: 同一位点不同深度对照组与模型组动物相比纹状体细胞外液中DA、DOPAC和HVA浓度均明显降低(P<0.01),且降低值均大于50%;而不同位点同一深度对照组与模型组动物相比纹状体细胞外液中DA、DOPAC和HVA浓度均明显降低小于50%,且在实验过程中出现死亡。 结论: 右侧同一位点不同深度注射6-OHDA模型成功率大于不同位点同一深度模型,且操作简单死亡率低。

Objective: To apply high efficiency liquid chromatography-electrochemistry(HPLC-ECD) method to measure monoamine neurotransmitters and their metabolites in rat brain microdialysate,and test the success rate for two kinds of Parkinson's disease (PD) rat model. Methods: The PD rat model was made with 6-hydroxydopamine (6-OHDA) injected either in different depths at the same site or in the same depth at different sites at the right side.The probe was inserted into the right striatum in rats at 7 days after operation.Ringer's solution was perfused in the speed of 1.5 μL·min-1 under conscious and free moving state.The dialysate was collected for one tube every 20 minutes and injected into the system of HPLC-ECD and dopamine (DA),3,4-dihydroxy acid (DOPAC),and homovanillic acid (HVA) contained in the dialysate were determined. Results: DA,DOPAC and HVA levels in striatal extracellular fluid were significantly reduced in the model group compared to the control group in different depths at the same site (P<0.01),and the reduction was greater than 50%;while the reduction of DA,DOPAC and HVA levels in striatal extracellular fluid in the model group compared to the control group in the same depth at different sites was significantly less than 50%,and death occurred during the trial. Conclusion: The success rate in the 6-OHDA injection model in different depths at the same site on the right side is higher than in the model in the same depth at different sites and the operation is simple with low mortality.
