目的:探讨猴鼻腔一致性切片制作方法,用于临床前鼻腔给药、吸入途径或其他途径给药药物的猴鼻腔组织病理学检查。方法:通过对食蟹猴鼻腔的解剖、固定、脱钙后,着重探讨了用于鼻腔给药药物及吸入毒性研究的猴鼻腔毒性病理检查一致性鼻腔标本4个切面的取材方法及参考的大体解剖标志,第1个切面在第1前磨牙前;第2个切面在第1、第2前磨牙之间;第3个切面在第1磨牙后,包括鼻腔旁边的上颌窦;第4个切面在鼻咽部软腭的前端和后端中间的位置。同时探讨了猴鼻腔组织HE染色切片制片中的其它注意事项。结果:制作出符合毒性病理学检查的猴鼻腔4个切面的HE染色切片,并清楚地显示了不同鼻粘膜上皮,包括鳞状上皮、移行上皮、嗅上皮和呼吸上皮。结论:本文描述了猴鼻腔毒性病理学检查的标准制片方法,运用本制片方法可以很好地制作出符合猴鼻腔毒性病理学镜检要求的一致性切片,用于临床前鼻腔给药药物、吸入途径或其它给药途径给药研究的猴鼻腔组织病理学检查。

Objective: To develop a method for preparing consistent slides of nasal cavity for histopathological examination of nasal biopsy from monkeys that received drug administration via nasal cavity,inhalation route or other route studies. Methods: The nasal cavities of young cynomolgus monkeys were necropsied,fixed, decalcified before tissue trimming.The trimming methods using the anatomic landmarks for the four levels of nasal cavity for the toxicological pathology examination of drugs administrated via nasal cavity or inhalation route were investigated in detail.The first level was the section anterior to the first premolar.The second level was the section between the first and second premolars.The third level was the section posterior to the first molars including the maxillary sinus adjacent to the nasal cavity.The forth level is section at the midway between the anterior and posterior end of the soft palate.Moreover, considerations for preparing HE stained slides were also discussed. Results: The HE stained tissue slides prepared at four different levels were consistent with the requirement of toxicological pathology examination.Four different epithelia of the nasal cavity,such as squamous epithelium,transitional epithelium,olfactory epithelium,and respiratory epithelium were also observed. Conclusion: The slide preparing method described in this paper can produce consistent slides that meet the requirement of toxicological pathology examination of monkey nasal cavity;it can be used for preclinical safety evaluation of these drugs administered via nasal cavity,inhalation route or other route studies.
