目的:建立同时测定雪松松针中丹皮酚、蛇床子素、和厚朴酚、厚朴酚4个有效成分含量的反相高效液相色谱法。方法:采用Agilent Eclipse plus-C18色谱柱(250 mm×4.6 mm,5 μm),以甲醇(A)-水(B)为流动相,梯度洗脱(0~7 min,55%A;7~20 min,80%A;20~25 min,55%A),流速1.0 mL·min-1,检测波长294 nm,柱温25 ℃。 结果:雪松松针中丹皮酚、蛇床子素、和厚朴酚、厚朴酚浓度分别在12.5~125、5~50、7~70和10~100 μg·mL-1范围内呈良好线性关系,相关系数在0.9989~0.9998之间;加样回收率(n=9)范围为98.6%~99.2%。结论:该方法简单、快速、高效,可作为雪松松针质量控制的参考方法。

Objective: To develop a RP-HPLC method for the simultaneous determination of four active constituents including paeonol, osthole,honokiol and magnolol in pine needles of Cedrus deodara. Methods: The Agilent TC-C18 column(250 mm×4.6 mm,5 μm)was adopted.The mobile phase was methanol(A)-water(B)with the gradient elution(0-7 min,55%A;7-20 min,80%A;20-25 min,55%A)at the flow rate of 1.0 mL·min-1.The column temperature was 25 ℃ and the detection wavelength was 294 nm. Results: Calibration curves were found to be linear in the ranges of 12.5-125 μg·mL-1 for paeonol,5-50 μg·mL-1 for osthole,7-70 μg·mL-1 for honokiol,and 10-100 μg·mL-1 for magnolol.The correlation coefficients were between 0.9989-0.9998.The average recoveries(n=9)of four active constituents were 98.6%-99.2%. Conclusion: The developed method is simple,rapid and efficient,which can be used for quality control of pine needles of C.deodara.
