在线固相萃取-HPLC测定复方甘草片中吗啡的含量

目的: 建立在线固相萃取(Online SPE)结合HPLC测定复方甘草片中吗啡含量的方法。方法: 采用Thermo Fisher Ultimate 3000双三元梯度液相色谱，以Acclaim C₁₈(4.3 mm×10 mm，5 μm)小柱对样品进行前处理和在线富集，后转载到分析柱Acclaim C₁₈(4.6 mm×250 mm，5 μm)上进行分离检测，流速为1.0 mL·min^-1，检测波长220 nm。结果: 吗啡在0.0239~0.239 mg·mL^-1(r=0.999 9)范围内线性关系良好，平均加样回收率为99.19%。结论: 该方法简便快捷，重现性好，节省大量时间与成本，方法精密度和准确度都有所提高，可为复方甘草片的质量控制提供方法。

Objective: To establish an HPLC method with online solid-phase extraction for the content determination of morphine in compound liquorice tablets. Methods: The analysis was carried out using Thermo Fisher Ultimate 3000 DGLC,an Acclaim C₁₈ column (4.3 mm×10 mm,5 μm) was used for sample pretreatment and online enrichment ,then the sample was separated and analyzed on an Acclaim C₁₈ column (4.6 mm×250 mm,5 μm) .The flow rate was 1.0 mL·min^-1 and the detective wavelength was 220 nm.Conclusion: Morphine showed a good linearity relationship in the range of 0.0239-0.239 mg·mL^-1,r=0.9999.The average recovery was 99.19%.The method is simple,fast,and reproducibe,with increasing precision and accuracy,which can be used for quality control of compound liquorice tablets.
