目的: 考察天麻素对大鼠脑内乙酰胆碱释放的影响。 方法: 大鼠皮下给药后利用脑内微透析技术进行连续动态取样,建立高效液相色谱-串联四极杆质谱定量分析方法测定脑透析液中的乙酰胆碱含量,考察不同剂量给药前后大鼠脑内不同部位乙酰胆碱释放量的变化。 结果: 实验发现天麻素皮下给药能剂量相关地增加大鼠脑内不同脑区中乙酰胆碱的释放。 结论: 天麻素的脑内作用机制可能与其对胆碱能神经递质释放的调节作用有关。

Objective: To study the effect of gastrodin on acetylcholine release in rat brain. Methods: A liquid chromatography-tandem mass spectrometry method was developed and validated for the determination of acetylcholine in rat brain dialysates.The dialysates were prepared by microdialysis before and after subcutaneous administration of gastrodin at different dosages. Results: The results indicated that gastrodin significantly increased acetylcholine release in rat medial prefrontal cortex,nucleus accumbens and hippocampus in a dose-related manner. Conclusion: The mechanism of gastrodins action in the brain may be related to the regulation of cholinergic neurotransmitter release.
