UPLC、HPLC测定血栓通注射液中5个皂苷类成分含量的比较研究

目的: 对比超高效液相色谱法与高效液相色谱法测定血栓通注射液中5个皂苷类成分(三七皂苷R1、人参皂苷Rg1、人参皂苷Re、人参皂苷Rb1、人参皂苷Rd)含量结果。 方法: 建立超高效液相色谱法测定血栓通注射液中5个皂苷类成分的测定方法,采用BEH C18色谱柱(2.1 mm×100 mm,1.7 μm),以乙腈(A)-水(B)为流动相梯度洗脱(0~8 min,20%A→22%A;8~9 min,22%A→40%A;9~13 min,40%A→55%A),检测波长203 nm,流速0.4 mL·min-1,柱温30 ℃。将测定结果与国家药品标准中高效液相色谱法结果进行比对。 结果: 超高效液相色谱仅15 min即可完成测试,测定时间远低于高效液相色谱法,2种方法所得结果一致。 结论: 超高效液相色谱法更加快速、高效。

Objective: To compare the ultra performance liquid chromatography(UPLC) and high performance liquid chromatography(HPLC) methods for determination of five saponins(notoginsenoside R1,ginsenoside Rg1,ginsenoside Re,ginsenoside Rb1,ginsenoside Rd) in Xueshuantong injection. Methods: A gradient UPLC system was used in the quantitative analysis on the reversed-phase column of Waters Acquity BEH(2.1 mm×100 mm,1.7 μm),using acetonitrile(A)-water(B) as the mobile phase(0-8 min,20%A→22%A;8-9 min,22%A→40%A;9-13 min,40%A→55%A),with a flow rate of 0.4 mL·min-1,the detective wavelength was set at 203 nm and the column temperature was 30 ℃.The five saponins were determined and compared by HPLC and UPLC,respectively. Results: The analysis time of UPLC was only 15 min which was obviously shorter than that of HPLC.Both of the two methods for the determination had the same results. Conclusion: UPLC is more rapid and effective than HPLC.
