目的: 利用高效液相色谱-质谱联用仪建立人血浆中地芬尼多的定性定量方法。 方法: 应用OasisTM HLB小柱进行固相萃取法提取,XBridge RP-18色谱柱分离。 结果: 在该条件下,人血浆中地芬尼多的线性范围为1.0~100.0 ng·mL-1,最小检出限为0.5 ng·mL-1。地芬尼多在1.0~100.0 ng·mL-1浓度范围内的回收率均在80%以上。 结论: 本研究所建立的方法适用于血浆中地芬尼多的定性定量检验。

Objective: To develop a qualitative and quantitative method by LC-MS/MS for the determination of difenidol in human blood. Methods: Difenidol was extracted from serum by OasisTM cartridges and separated on the XBridge RP-18 column. Results: Under this condition,the calibration curve of difenidol in human plasma was linear within the injection amount ranging from 1.0 ng·mL-1 to 100 ng·mL-1,the minimum limit of detection was 0.5 ng·mL-1 and the recovery was above 80% for difenidol within the range of 1.0-100 ng·mL-1. Conclusion: The described LC-MS/MS method can be applied to the qualitative and quantitative determination of difenidol in human plasma.
