高效液相色谱法同时测定减肥类保健食品中盐酸西布曲明和酚酞

目的: 采用高效液相色谱(HPLC)建立了减肥类保健食品中盐酸西布曲明和酚酞的同时测定方法。 方法: 以乙腈提取样品中盐酸西布曲明和酚酞,测定时以Cloversil-C₁₈(4.6 mm×150 mm,5 μm)为分析柱,乙腈-己烷磺酸钠柠檬酸溶液(60: 40,v/v)为流动相,流速1.0 mL·min^-1,检测波长为223 nm。 结果: 盐酸西布曲明和酚酞在0.001~0.1 mg·L^-1范围内线性关系良好,相关系数(r)大于0.9993,盐酸西布曲明和酚酞最低检出浓度分别为5 mg·kg^-1和2.5 mg·kg^-1。减肥茶、减肥胶囊和减肥片的5.0,10,50 mg·kg^-1的平均添加回收率均大于80%, RSD(n=6)小于10%。 结论: 本方法简便快速、灵敏度高、结果准确可靠,可实现2种成分同时测定,适用于减肥类保健品中盐酸西布曲明和酚酞的监督检验。

Objective: To develop a method for simultaneous determination of sibutramine hydrochloride and phendphthaein in weight loss functional foods with high performance liquid chromatography. Methods: Sibutramine hydrochloride and phendphthaein were extracted by acetonitrile from samples and separated by the column of Cloversil-C₁₈(4.6 mm×150 mm,5 μm) with mobile phase solution of acetonitrile and sodium hexanesulfonate and citric acid(60: 40,v/v). The flow rate is 1.0 mL·min^-1 and the determination wavelength was 223 nm. Results: The linear range for sibutramine hydrochloride and phendphthaein were from 0.001 mg·L^-1 to 0.1 mg·L^-1 and the correlation coefficient(r) was above 0.9993. The determined limits of sibutramine hydrochloride and phendphthaein were 5.0 mg·kg^-1and 2.5 mg·kg^-1.The average recoveries were above 80% at spike levels of 5.0 mg·kg^-1, 10.0 mg·kg^-1 and 50.0 mg·kg^-1 in slimming tea, capsule and tablet. And the relative standard deviations of 6 repeated spiking experiments were below 10%. Conclusion: As a rapid, easy and accurate method, it can simultaneously determine the two additive drugs, and was suitable for determining these two additive drugs in weight loss functional foods.
