目的:建立同时测定莱阳梨清膏中3个咖啡酰奎宁酸类成分3-O-咖啡酰奎宁酸、绿原酸(5-O-咖啡酰奎宁酸)、4-O-咖啡酰奎宁酸的HPLC方法。方法:采用Thermo BDS(4.6 mm×250 mm,5 μm)色谱柱,以乙腈(A)-0.4%磷酸溶液(B)为流动相,梯度洗脱(0~35 min,5%A→10%A;35~40 min,10%A),流速1.0 mL·min-1,检测波长为327 nm。结果:3-O-咖啡酰奎宁酸、绿原酸(5-O-咖啡酰奎宁酸)、4-O-咖啡酰奎宁酸进样量分别在0.023~0.230,0.022~0.224,0.023~0.226 μg范围内与峰面积呈良好的线性关系;平均回收率(n=6)分别为97.2%,97.0%,99.5%。结论:本法操作简便,结果准确,灵敏度高,可用于莱阳梨清膏中多指标成分的质量控制。

Objective:To develop an HPLC method for simultaneously determination of three caffeoylquinic acids(3-O-caffeoylquinic acid,chlorogenic acid,4-O-caffeoylquinic acid)in Laiyang pear clear paste. Methods:Thermo BDS(4.6 mm×250 mm,5 μm)column was adopted.The mobile phase consisting of acetonitrile(A)-0.4% phosphoric acid solution(B)with gradient elution(0-35 min,5%A→10%A;35-40 min,10%A)was applied at a flow rate of 1.0 mL·min-1.The detection wavelength was 327 nm. Results:The calibration curves were linear in the ranges of 0.023-0.230,0.022-0.224,0.023-0.226 μg for 3-O-caffeoylquinic acid,chlorogenic acid(5-O-caffeoylquinic acid),4-O-caffeoylquinic acid;The average recoveries(n=6)were 97.2%,97.0% and 99.5%,respectively. Conclusions:The method is simple,accurate,and sensitive,which can be used for quality control of multi-component in Laiyang pear clear paste .
