目的:建立同时测定肉豆蔻中丁香酚、异丁香酚、肉豆蔻醚和榄香脂素4个成分含量的气相色谱法。方法:样品用乙酸乙酯超声提取,采用GC法对肉豆蔻中丁香酚、异丁香酚、肉豆蔻醚和榄香脂素进行含量测定。气相色谱条件:HP-5 MS石英毛细管柱(0.25 mm×30 m×0.25 μm);氢火焰离子化检测器(FID);进样口温度240℃;检测器温度250℃;柱温起始温度为40℃,保持1 min,以5℃·min-1的速率升高到220℃,保持1 min;载气为氮气,流速1 mL·min-1,分流比为10:1;进样量1 μL。结果:丁香酚、异丁香酚、肉豆蔻醚和榄香脂素4个成分在相应的浓度范围内呈现良好的线性关系;加样回收率(n=9)分别为99.4%,99.3%,98.2%,98.8%;肉豆蔻不同部位中丁香酚、异丁香酚、肉豆蔻醚和榄香脂素的含量有明显差异。结论:本法简便快速,可以用于肉豆蔻中丁香酚、异丁香酚、肉豆蔻醚和榄香脂素4个成分的含量测定。

Objective:To establish a GC method for simultaneous determination of four components (eugenol, isoeugenol, myristicin, elemicin) in Myristica fragrans . Methods:Samples were extracted with ethyl acetate using supersonic fluid extraction. GC method was applied for the determination of eugenol, isoeugenol, myristicin and elemicin in Myristica fragrans . Chromatographic conditions:HP-5MS capillary column(0.25 mm×30 m×0.25 μm) was adopted;the detector was FID;the inlet temperature was 240℃;the detector temperature was 250℃;the column temperature was programmed to start at 40℃, rising to 220℃ at a rate of 5℃·min-1. Nitrogen(1 mL·min-1) was used as the carrying gas, and the split ratio was 10:1;the injection volume was 1 μL. Results:Four components(eugenol, isoeugenol, myristicin, elemicin) had good linear relationship in the corresponding concentration ranges. The average recoveries(n=9) of the above four components were 99.4%, 99.3%, 98.2%, and 98.8%, respectively. The contents of four components in different parts of Myristica fragrans were different. Conclusions:This method is simple, quick and specific, which can be applied for the simultaneous determination of eugenol, isoeugenol, myristicin, elemicin in Myristica fragrans .
