目的: 研究建立拉曼光谱法分析验证氢氯噻嗪包合物的可行性。 方法: 分别采用拉曼光谱法和傅里叶红外光谱法测定并获得β-CD、氢氯噻嗪、β-CD和氢氯噻嗪物理混合物、氢氯噻嗪包合物四种物质固体粉末的拉曼图谱和红外图谱,并将其进行对照分析。 结果: 拉曼光谱可得到与红外光谱一致的结果,表明氢氯噻嗪以氢键作用嵌入β-环糊精的疏水空腔形成了包合物,从而增加了氢氯噻嗪的稳定性和溶解度。 结论: 拉曼光谱法具有快速、准确、简便、无损测量等显著优势,可作为验证药物包合物的形成的手段。

Objective: To study the feasibility to identify the formation of hydrochlorothiazide-β-cyclodextrin inclusion compound by Raman spectroscopy. Methods: The Raman spectrum and Infrared spectrum of hydrochlorothiazide,β-cyclodextrin,β-cyclodextrin and hydrochlorothiazide physical mixture,hydrochlorothiazide inclusion compound in the solid state are obtained respectively used confocal Raman spectrometer and Fourier transform Infrared spectrometer,then compared the relevant spectrums to reveal the structural features. Results: The Raman spectrum indicated that the hydrochlorothiazide inclusion compound is formed by hydrogen bonds between hydrochlorothiazide and β-cyclodextrin,which is in accordance with the conclusion obtained in the infrared spectrum. Conclusion: Raman spectroscopy has significant advantages as a rapid,accurate,simple,non-destructive measurement and can be used as an important method to validate the formation of inclusion compound.
