玉屏风颗粒2个异黄酮类及4个色原酮类成分的含量测定

目的: 建立同时测定玉屏风颗粒中2个异黄酮类成分(毛蕊异黄酮葡萄糖苷、毛蕊异黄酮)及4个色原酮类成分(升麻素苷、升麻素、5-O-甲基维斯阿米醇苷、亥茅酚苷)含量的高效液相色谱法。 方法: 采用INERSTIL ODS3(4.6 mm×250 mm,5 μm)色谱柱,以乙腈-0.2%磷酸溶液为流动相,梯度洗脱,流速1.0 mL·min-1,检测波长为254 nm。 结果: 升麻素苷、毛蕊异黄酮葡萄糖苷、升麻素、5-O-甲基维斯阿米醇苷、亥茅酚苷、毛蕊异黄酮线性范围分别为5.3~263.6,2.1~103.6,7.5~376.2,7.4~396.8,2.0~98.8,4.4~220.6 μg·mL-1(r≥0.9999);平均回收率(n=6)分别为99.9%,95.1%,95.7%,96.8%,101.7%,99.0%。 结论: 该法准确、可靠,可用于玉屏风颗粒的质量控制。

Objective: To develop an HPLC method for simultaneous determination of two isoflavonoids,(calycosin-7-O-β-D-glucoside,calycosin) and four chromones(prim-O-glucosylcimifugin,cimifugin,4'-O-β-glucopyranosyl-5-O-methylvisamminol,sec-O-glucosylhamaudol) in Yupingfeng granules. Methods: The six components were separated by INERSTIL ODS3(4.6 mm×250 mm,5 μm) column with gradient elution of acetonitrile-0.2% phosphoric acid as the mobile phase,the flow rate was 1.0 mL·min-1,and the detection wavelength was 254 nm. Results: The calibration curves were linear in the ranges of 5.3-263.6 μg·mL-1 for prim-O-glucosylcimifugin,2.1-103.6 μg·mL-1 for calycosin-7-O-β-D-glucoside,7.5-376.2 μg·mL-1 for cimifugin,7.4-396.8 μg·mL-1 for 4'-O-β-glucopyranosyl-5-O-methylvisamminol,2.0-98.8 μg·mL-1 for sec-O-glucosylhamaudol,4.4-220.6 μg·mL-1 for calycosin(r≥0.9999).The average recoveries(n=6) were 99.9%,95.1%,95.7%,96.8%,101.7%,99.0%,respectively. Conclusion: This method is simple,rapid and accurate with good reproducibility,it is suitable for the quality control of Yupingfeng granules.
