紫草中羟基萘醌总色素的分光光度定量方法研究

目的: 探讨紫草中的羟基萘醌总色素的定量分析方法。 方法: 分别以紫草中的6个羟基萘醌类色素成分紫草素(SK)、去氧紫草素(DOSK)、 β、 β-二甲基丙烯酰基紫草素(DASK)、异丁酰基紫草(IBSK)、异戊酰基紫草素(IVSK)和2-甲基正丁酰基紫草素(MNBSK)为标准,建立紫草中羟基萘醌总色素的不同分析方法,并采用统计分析手段与中国药典收录的传统方法进行比较。 结果: 6种标准对照物均可建立线性范围良好的标准曲线,SK、DOSK、DASK、IBSK、IVSK和MNBSK的相关系数依次为0.9997,0.9989,0.9998,0.9995,0.9999,0.9997,线性范围依次为5.6~106.0 μg·mL-1,5.3~106.0 μg·mL-1,5.9~117.0 μg·mL-1,5.6~112.0 μg·mL-1,5.2~105.0 μg·mL-1和5.6~111.0 μg·mL-1;药典的传统方法与以SK和DOSK为标准的测定结果差异不明显,但与其余4种物质为标准的测定结果差异极为显著,且前者对后者的定量误差较大。 结论: 以SK为标准的标准曲线法和药典收录的传统公式计算法对紫草中的羟基萘醌类总色素含量的测定结果并无差异,且公式计算法简单、方便,适合作为监控手段。但由于紫草中羟基萘醌类总色素以DASK和IVSK的相对含量最高,因此,以DASK为标准定量紫草中萘醌类总色素更为准确。

Objective: To discuss the determinative methods for hydroxynaphthoquinone pigments derive from Zicao . Methods: Six methods were established for determination total hydroxynaphthoquinone pigments in Arnebia euchroma (Royle) Johnst by using with respective standard references,namely shikonin (SK),deoxyshikonin (DOSK),Isobutyrylshikonin (IBSK),β,β-dimethylacrylshikonin (DASK),isovalerylshikonin (IVSK) and 2-Methyl-n-butylshikonin (MNBSK).And a statistical analysis method was used in comparing results with the method of Chinese Pharmacopoeia. Results: The results showed that the calibration curves of six shikonin derivatives had a good linear relation.The correlation coefficients R were 0.9997,0.9989,0.9998,0.9995,0.9999,0.9997 and the linearity ranges were 5.6-106.0 μg·mL-1,5.3-106.0 μg·mL-1,5.9-117.0 μg·mL-1,5.6-112.0 μg·mL-1,5.2-105.0 μg·mL-1,5.6-111.0 μg·mL-1 for SK,DOSK,DASK,IBSK,IVSK,MNBSK,respectively.There is not significant difference between the method of Chinese Pharmacopoeia and take SK and DOSK as a standard,but it have a significant difference above-mentioned with others shikonin derivatives. Conclusions: There was no significant difference between the method of Chinese Pharmacopoeia and standard calibration curve method which take SK as a standard for the contents of hydroxynaphthoquinone pigments,and the formulas method of Chinese Pharmacopoeia have the merit such as simple,rapid,and easy to realize.However,DASK and IVSK was the main components in total hydroxynaphthoquinone pigments of Arnebia euchroma (Royle) Johnst,so it will be much more accurate for quantitative methods of total hydroxynaphthoquinone pigments with DASK as a standard.
