高效液相色谱法测定对硝基苯-α-D-吡喃葡糖苷的含量

目的: 建立高效液相色谱法测定对硝基苯-α-D-吡喃葡糖苷的含量。 方法: 采用Hypersil C₁₈ 色谱柱(250 mm×4.6 mm,5 μm);流动相-甲醇-水(759∶ 25),流速为0.8 mL·min^-1,紫外检测器:检测波长 303 nm,柱温为35 ℃。 结果: 对硝基苯-α-D-吡喃葡糖苷的浓度在 13.5~135.0 μg·mL^-1 的范围内,峰面积与浓度呈良好线性(r=0.9999)。方法平均回收率为 99.7%(n=6)。 结论: 本法简便、快速、准确,可用于对硝基苯-α-D-吡喃葡糖苷的含量测定。

Objective: To establish an HPLC method for determination of P-nitrophenly-α-D-glucopyranoside(α-PNPG). Methods: α-PNPG was determinated by HPLC with a Hypersil C₁₈(column,250 mm×4.6 mm,5 μm)with UV detection at 303 nm,the mobile phase was contained of methanol-water(75∶ 25),the flow rate was 0.8 mL·min^-1,the column temperature was 35 ℃. Results: The calibration curve was linear in the range of 13.5-135.0 μg·mL^-1 (r=0.9999) for α-PNPG.The average recovery was 99.7% (n=6). Conclusion: The method is simple,rapid,accurate ,reliable,high sensitivity and good separation ,which may be used for the determination of α-PNPG.
