目的: 建立HPLC法测定苦豆子中野靛碱和槐胺碱的含量。 方法: 采用Waters x-Bridge C18(4.6 mm×250 mm,5 μm)色谱柱,流动相为乙腈-含2.0 mL·L-1三乙胺的0.05 mol·L-1的磷酸二氢钾溶液(90∶ 10),流速为1.0 mL·min-1,检测波长为311 nm。 结果: 野靛碱和槐胺碱的线性范围分别为4.972~165.7 μg(r=0.9994)和4.388~146.3 μg(r=0.9991);平均回收率(n=9)分别为101.8%和99.3%。 结论: 本方法简便、可靠、准确,可为苦豆子药材的开发利用提供理论依据。

Objective: To develop an HPLC method for determination of cytisine and sophoridine in Sophora alopecuroides. Methods: Waters x-Bridge C18(4.6 mm×250 mm,5 μm) column was adoped;The mobile phase consisted of acetonitrile-0.05 mol·L-1 potassium dihydrogen phosphate solution containing 2.0 mL·L-1 triethylamine(90∶ 10) at the flow rate of 1 mL·min-1.The detection wavelength was set at 311 nm. Results: The linear ranges of cytisine and sophoridine were 4.972-165.7 μg(r=0.9994) and 4.388-146.3 μg(r=0.9991);The recoveries of them were 101.8% and 99.3%. Conclusion :The method is simple,reliable,accurate,and can provide basis for development and utilization of Sophora alopecuroides.
