目的: 建立川芎中川芎嗪的含量测定方法,并测定川产道地药材川芎中川芎嗪的含量。 方法: 采用高效液相色谱法测定川芎中川芎嗪的含量。色谱柱:Phenomenex Gemini C18(250 mm×4.6 mm,5 μm);流动相:甲醇-四氢呋喃-20 mmol·L-1醋酸铵水溶液(pH 9.5)(68∶ 7∶ 425);流速:1.0 mL·min-1;检测波长:280 nm;柱温:35 ℃。 结果: 川产道地药材川芎中川芎嗪含量甚微,大多数药材含量低于检测限0.10 μg·g-1而未检出。 结论: 川芎嗪为川芎中主要有效成分是一个认识误区,本方法测定结果可靠,为进一步科学认识川芎药效成分提供参考。

Objective: To establish determination method of tetramethylpyrazine in Ligusticum chuanxiong Hort.,and determine the content of tetramethylpyrazine in geo-authentic Ligusticum chuanxiong Hort. of Sichuan province. Methods: The contents of tetramethylpyrazine were determined by HPLC.HPLC was carried out with a Phenomenex Gemini C18(250 mm×4.6 mm,5 μm) column,the mobile phase was methanol-tetrahydrofuran(THF)-20 mmol·L-1 ammonium acetate water solution(pH 9.5)(68∶ 7∶ 425) with a flow rate of 1.0 mL·min-1,the detection wavelength was 280 nm and the column temperature was 35 ℃. Results: The content of tetramethylpyrazine in geo-authentic Ligusticum chuanxiong Hort. of Sichuan province was minimal,and mostly lower than 0.10 μg·g-1 then not be detected. Conclusion: There is a cognition mistake that tetramethylpyrazine is the main active component in Ligusticum chuanxiong Hort..The result determined by this method is reliable, which provide the basis in order to further scientific cognition about pharmacodynamic action component of Ligusticum chuanxiong Hort.
