全蝎蛋白药效组分对HepG2细胞的体外抑制作用

目的: 研究全蝎(Scorpio)蛋白药效组分对HepG2细胞的体外抑制作用,为建立全蝎质量生物效应评价方法和标准奠定基础。 方法: 采用改良MTT法、流式细胞法研究全蝎蛋白药效组分对HepG2细胞毒和细胞周期抑制的作用。 结果: 抑制癌细胞生长的浓度大于或等于37 mg·mL-1,抑制率与浓度呈正比,相关系数为0.9157,IC50为244 mg·mL-1,显效时间0~48 h;全蝎蛋白药效组分浓度大于或等于9.25 mg·mL-1时,能显著提高HepG2细胞的凋亡率。 结论: 全蝎蛋白药效组分具有促进HepG2细胞凋亡、抑制增殖的作用,其生物效应指标为9.25~175 mg·mL-1,可作为全蝎质量生物效应评价指标之一。

Objective: To investigate the inhibitory effect of protein active composition of Scorpio on human hepatocarcinoma cell line HepG2 in vitro,and to supply the data on establishing the quality evaluation method of biological effect guided by the clinical effect of Scorpio. Methods: HepG2 cells were examined by trypan blue exclusion.The proliferation of cells was determined by improving MTT assay.Flow cytometry was performed to analyze cell apoptosis with propidium iodide(PI). Results: When the concentration of protein active composition of Scorpio exceeded or equaled 37 mg·mL-1,the coefficient correlation of the growth inhibiting curve to HepG2 cells was 0.9157,and IC50 was 244 mg·mL-1.The excellence time was 0 to 48 hours.When the concentration of protein active composition of Scorpio exceeded or equaled 9.25 mg·mL-1,the apoptosis ratio of HepG2 cells was raised significantly. Conclusion: The protein active composition of Scorpio might promote the apoptosis of HepG2 cells and restrain the proliferation of them.The antitumor biological effect of the protein active composition of Scorpio was 9.25-175 mg ·mL-1. The protein active composition may be one of the indexes for quality evaluation of biological effect of Scorpio.
