青天葵野生球茎和组培球茎中氨基酸的分析

目的: 测定青天葵野生球茎和组培球茎中氨基酸含量。方法:采用AccQ-Tag法,以AccQ-FlourTM对青天葵野生球茎和组培球茎中17种氨基酸进行柱前衍生;采用Waters 1525HPLC仪器、AccQ-TagTM柱,以AccQ·Tag浓液-水(1∶10)为流动相A,乙腈-水(60∶40)为流动相B进行梯度洗脱,检测波长248 nm,柱温37 ℃,进样量10 μL。 结果:[ 17种氨基酸的线性回归方程r值均>0.9997,平均回收率为98.7%,RSD为1.8%(n=6);青天葵野生球茎和组培球茎中均含有17种氨基酸,组培球茎中的总氨基酸含量高于野生球茎,但二者差异无统计学意义(P>0.05)。 结论: 本法快速、简便,结果准确;从氨基酸含量来考察,组织培养的青天葵具有与野生青天葵相同的品质。

Objective: To determine the contents of amino acids analysis of amino wild corm and tissue culture corm of Nervilia fordii. Methods: Amino acids in the injections were pretreated with AccQ-FlourTM separately and their derivatives were analyzed on Waters 1525 HPLC using AccQ-Tag column.Mobile phase consisted of AccQ·Tag concentrated liquid-water(1∶ 10)(A) and acetonitrile -water(60∶ 40)(B) with gradient elution.The detection wavelength was 248 nm,column temperature 37 ℃,and injection volume was 10 μL. Results: The linear regression equation was greater than 0.9997 in 17 amino acids,the average recovery was 98.7%,the RSD was 1.8% (n=6).17 amino acids of wild corm and tissue culture corm were detected,the total content of amino acids of tissue culture corm was higher than that of wild corm,but the difference was no significant (P>0.05). Conclusion: This method is rapid,simple and accurate.The wild and tissue culture of Nervilia fordii have the same quality according to contents of amino acids.
